Objective:Through analysis methods such as Amplitude of Low Frequency Fluctuation(ALFF),fractional Amplitude of Low Frequency Fluctuation(FALFF)and Regional Homogeneity(Re Ho)based on SPM softwares,to explore the correlation between negative life events and brain functional activities of first-episode adolescent Major Depressive Disorder(MDD),so as to provide basis for further exploration of the etiology and intervention measures of first-episode adolescent depression.Methods: From November 2018 to May 2020,54 first-episode adolescent MDD patients(as the MDD group)accepted in sichuan province center for mental health clinic outpatient and hospitalization and 30 recruited adolescents(as the HC group)matched by gender,age and educational background recruited were assessed by the Hamilton Anxiety Scale(HAMA),the Hamilton Depression rating Scale-24(HAMD-24)and the Adolescent Self-Rating Life Events Check List(ASLEC).In addition,resting-state f MRI scan was also performed on both groups.SPSS22.0 software was used for statistical analysis of demographic data and clinical symptom scale results.Two-sample t-test and chi-square test were used to compare general data and scales between the two groups.ALFF,FALFF,REHO and other analysis methods based on SPM software were used to compare brain functional activities between the MDD group and HC group,and the brain regions with differences between the two groups were obtained.The possible influencing factors such as HAMD-24 totals scores were used as covariates to analyze the correlation between different brain regions of the two groups and total score and factor scores of ASLEC in MDD group.Results: 1.Comparison of clinical data: the HAMD-24,HAMA,ASLEC total scores and each individual factor scores of MDD were higher than those of HC(P all <0.001).2.Comparison of ALFF,f ALFF and Re Ho values between the two groups: In contrast with HC group,ALFF values of MDD group in bilateral thalamus,right fusiform gyrus and bilateral parhippocampal gyrus were increased(P<0.01).Comparied with HC group,f ALFF value of MDD group in cuneus,medial Prefrontal Cortex(m PFC)and cerebellum was increased(P<0.01).In contrast with HC group,the Re Ho value of MDD group in the talar gyrus and right cerebellum was increased(P<0.01).3.Correlation analysis: For MDD group,the total scorse of ASLEC,interpersonal factor and other factors were positively correlated with total score of HAMD-24(r=0.286~ 0.320,P<0.05).4.Correlation analysis of ALFF,f ALFF,Re Ho and HAMD-24 total scores between the two groups: The ALFF of left parhippocampal gyrus and the f ALFF of the cuneus were negatively correlated with HAMD-24 total scores(r=-0.430~-0.373,P< 0.05).5.The HAMD-24 total scores was used as the covariable,and the correlation analysis of ALFF,f ALFF,Re Ho and the total scores and factor scores of ASLEC in the two groups with different brain regions between the two groups was as follows: For ALFF value,bilateral ALFF value was positively correlated with the loss factor(r=0.286,P=0.044).In the terms of f ALFF value of MDD group,f ALFF value of m PFC was positively correlated with the ASLEC total scores,interpersonal relationship,learning stress and health adaptation factors(r= 0.293~0.336,P<0.05).For Reho value,Right cerebellum Reho value was negatively correlated with health adaptation factor(r=-0.317,P=0.028).Conclusion: 1.The more severe the influence degree of NLE in firstepisode adolescent MDD patients,the more severe the depression degree.2.Brain regions such as thalamus,temporal lobe,occipital lobe,medial prefrontal cortex,cerebellum and limbic system may be involved in the formation,control and expression of emotions in first-onset adolescent MDD patients.3.NLE is correlated with brain functional activities of thalamus and MPFC in firstepisode adolescent MDD patients,and NLE may be an important influencing factor for abnormal brain functional activities in first-episode adolescent MDD patients.
