Backgroud: Renal Cell Carcinoma(RCC),also known as Kidney cancer,is a common urologic malignancy.Kidney cancer has a high incidence and mortality rate,with an estimated 66,800 new cases and 23,400 deaths in China in 2015.Kidney cancer is not easily detected in the early stage,and about 20% to 30% of patients have already developed distant metastasis by the time of diagnosis,while patients with metastatic kidney cancer have a poor prognosis.This study focuses on analysis of von Willebrand factor(vWF)expression level in ccRCC,the relationship between vWF and prognosis of patients with ccRCC,as well as the potential mechanism of action,verify whether vWF can be used as a tumor molecular markers,help the early diagnosis and treatment of the disease,hope to provide new targets for targeted therapy drugs research and development.Methods: Using bioinformatics methods,rna-seq expression data and paired clinical information were downloaded from The Cancer Genome Atlas(TCGA)database,and ActivePerl and R software were used for data processing and statistical analysis.To analyze the expression level of vWF in renal clear cell carcinoma,to investigate the relationship between vWF and prognosis of patients with renal clear cell carcinoma,and to analyze the potential function and signaling mechanism of vWF in renal clear cell carcinoma by GSEA enrichment.Results : The expression of vWF in renal clear cell carcinoma tissues was significantly higher than that in adjacent tissues(P <0.001),and the mRNA expression level of vWF was closely related to clinical Stage,T Stage,M Stage,N Stage and pathological Grade(P <0.001).Overall survival(OS)was significantly higher in the high-expression VWF group than in the low-expression VWF group(P=0.004).After adjusting for related variables,multivariate Cox regression analysis showed a significant correlation between high vWF expression and good prognosis(HR: 0.996;95% CI: 0.994 ~ 0.999;P=0.009)correlation.Gene set enrichment analysis(GSEA)showed that vWF was associated with sprouting angiogenesis,proliferation and migration of endothelial cells,cell adhesion,KEGG renal cell carcinoma pathway,TGF-? signaling pathway,ERBB signaling pathway,mTOR signaling pathway and VEGF signaling pathway.Conclusion: 1.The expression level of vWF in ccRCC tumor tissues was significantly higher than that in adjacent tissues.2.High expression of vWF is associated with good prognosis in patients with renal clear cell carcinoma,but it is not considered that vWF can be used as a tumor molecular marker to judge prognosis.3.vWF may be related to sprouting angiogenesis,endothelial cell proliferation and migration,cell adhesion,KEGG renal cell carcinoma pathway,TGF-signaling pathway,ERBB signaling pathway,mTOR signaling pathway,VEGF signaling pathway.
