With the rapid pace of life and all kinds of pressure,fatigue has become a common symptom in people’s life.If the symptoms of fatigue can not be relieved for a long time,it will affect people’s work efficiency and quality of life,and even lead to physical or psychological diseases such as depression.Fatigue has become an increasing concern in the world.Ginseng root is rich in ginsenoside and ginseng polysaccharide,and jujube and lily are also rich in polysaccharide,which can safely and effectively prevent physical fatigue.Therefore,in this study,ginseng,jujube and lily were used to anti-fatigue granules.Firstly,the extraction process parameters were optimized,the molding process of granules was investigated,the quality of granules was studied,and the anti-fatigue effect was evaluated.Objective to study the extraction of effective components from traditional Chinese medicine.Taking the extraction rate and the total ginsenoside(Rg1+Re+Rb1)extraction amount as the evaluation indexes,the ethanol extraction process of ginsenoside was studied by central composite response surface methodology.The optimal ethanol extraction process parameters were determined as follows:the alcohol concentration was 65%,the solid-liquid ratio was 1:15,the extraction time was 2 times,1 hour each time.The water extraction method was used to extract the polysaccharide from the prescription and the single factor method was used to investigate the effects of solid-liquid ratio,extraction time and extraction temperature on the extraction rate and the amount of crude polysaccharide.According to the content of crude polysaccharide,the water extraction process parameters were determined as follows:the ratio of material to liquid was 1:15,the extraction temperature was 90℃,the extraction time was 2 h,and the extraction time was 2 times.The molding process of granules was studied.The types of excipients,the dosage of excipients and the concentration of wetting agent were investigated with the condition of granules,solubility,formability,fluidity and hygroscopicity as the indexes.The results showed that the mass ratio of main drug and excipient was 1:0.8,the excipient was a mixture of soluble starch and dextrin(7:3),and 93%ethanol was used as wetting agent.The quality of granules was studied.TLC was used for qualitative identification of jujube and lily,HPLC was used for quantitative determination of ginsenosides(Rg1+Re+Rb1)in granules,UV method was used for determination of crude polysaccharide,the specificity was good.There was negative interference in TLC identification of lily,so it was not included in the quality standard.TLC method was used to identify jujube granules.The test sample and the control medicine showed the same color spots at the corresponding position,so it was included in the quality standard.The specificity and durability of total ginsenosides in granules determined by HPLC were good.The method is simple and feasible.The routine inspection of granules,such as particle size,solubility,loading difference and water content,met the corresponding requirements.According to the content determination results,the content of ginsenoside(Rg1+Re+Rb1)in each gram of granules should not be less than 1.59 mg/g,and the content of crude polysaccharide should not be less than 294.52 mg/g.The anti-fatigue activity of the granules was evaluated by using the fatigue model of swimming mice.Kunming mice were randomly divided into low,medium and high dose groups and blank control group.The doses of low,medium and high dose groups were 0.83 g/kg/d,1.65 g/kg/d and 2.25 g/kg/d respectively.The blank dose group was given the same volume of normal saline,forced swimming test.The swimming time of mice in each group was recorded,and the contents of serum urea nitrogen(BUN),liver glycogen and muscle glycogen were determined after swimming.The results showed that the three doses of ginseng group granule could prolong the swimming time of mice with load,and there was statistical difference(*P<0.05).The middle and high dose group could significantly reduce the content of BUN in serum and increase the content of muscle glycogen and liver glycogen in mice(*P<0.05).The results showed that the ginseng granules prepared in this study had the effect of physical anti-fatigue,which provided experimental basis for the development and application of the compound health food.
