| Red ginseng is the dried root and rhizome of araliaceae ginseng(Panax ginseng C.A.Mey). Red ginseng is sweet and slightly bitter in flavour, which has the function of nourishing vitality, invigorating spleen and lung, promoting the secretion of body fluid and nourishing blood and so on. Praised as “ King of Herbsâ€, ginseng is not only medicine but also food, whose main bioactive component is ginsenoside. Argininyl fructoyl glucose(AFG) obtained from ginseng is a kind of amino acid derivative with pharmacological activity effects.Ginseng was made from red ginseng in the maillard reaction to produce a new no ginsenoside component. Researches have shown that AFG has the effects of promoting microcirculation, decreasing blood sugar, antioxidant and inhibiting melanin formation. Based on previous researches, the research gives a further and in-depth study in order to provide some implication for the application and development of no ginsenoside component in ginseng.In this experiment, red ginseng was used as experimental material and then was smashed. After filtration, extracting was added 10 times volume methanol for 40 min by ultrasonic extraction machine. Then, extraction was added 100 times volume water for 40 min again by ultrasonic extraction. Extracting solution was filtered with 1k Da hollow fiber membrane, filter liquor was made into Non-ginsenoside Argininyl Fructoyl Glucose(AFG) through vacuum freeze dryer. Test their purity by HPLC and used as further experiment drug.The mice was gavaged with AFG for 28 d, and then made an forced swimming test. In anti-fatigue experiment, result show that there are not significant difference in terms of weight and organ index in control group and treatment group; the force swimming time of treatment group is significantly increased; volume of lactic acid(LD), urea nitrogen(BUN) and malondialdehyde(MDA) in serum of treatment group are significantly reduced; volume of hepatic glycogen(Gly), glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) in serum of treatment group are significantly increased; volume of PGC-1α and PPAR-α in Gastrocnemius of treatment group are significantly increased. The results of research were consistent with the blotting. In addition, we make modle of mice with low immune function by injecting cyclophosphamide(CY). The mice was gavaged with AFG for 21 d. Findings show compared with model group, weight and organ index of mice in treatment group are significantly increased in immunity experiment. Volume of GSH-Px and SOD in serum of treatment group are significantly increased, while volume of MDA in serum of treatment group is significantly reduced. Volume of IL-2 and Ig G in serum of treatment group are significantly increased. Levels of CD3, CD4, CD8 in spleen lymphocyte and spleen lymphocyte proliferation and are significantly increased.To sum up,Non-ginsenoside Argininyl fructoyl glucose obtained from red ginseng has the effect of anti-fatigue and improving immunity in mice. |
