Background: Renal clear cell carcinoma is one of the most common malignant tumors in the urinary system.Its morbidity and mortality are high.Due to its hidden onset and unobvious clinical features,many patients are already at an advanced stage when they are diagnosed,and treatment is quite difficult.The 5-year survival rate is only 5-10%,and the prognosis is poor.G protein-coupled receptor kinase 6(GRK6)plays an important role in the occurrence,development and prognosis of various diseases,such as tumor progression,angiogenesis,cancer cell proliferation and metastasis.GRK6 is highly expressed in gastric cancer,colorectal cancer,liver cancer,thyroid cancer,neuroglioma and other cancers.It is an independent indicator of poor prognosis.GRK6 is also one of the key factors required for metastatic medulloblastoma.The proliferation and metastasis of lung cancer cells can be caused by the down-regulation of GRK6 expression in lung cancer,however Its expression level in renal clear cell carcinoma has not been clarified,and whether it plays a role in renal clear cell carcinoma is also unknown.The expression of GRK6 in renal clear cell carcinoma and its clinical significance were explored in this experiment through the application of public biological data platforms and experiments,and the experiment analyzes its possible molecular mechanism,and verifies whether it can be used as a novel biomarker,making it a potential target for targeted therapy.This will help the diagnosis,treatment and prognosis of renal clear cell carcinoma.Methods and Materials: We use Oncomine database and use UALCAN,c Bio Portal for Cancer Genomics,Kaplan-Meier Plotter platform and other bioinformatics methods to analyze the difference of GRK6 gene expression level in renal clear cell carcinoma and adjacent tissues and analyze the impact of GRK6 expression levels on the prognosis of patients in TCGA(The Cancer Genome Atlas,TCGA)database.According to the inclusion and exclusion criteria,30 cases of cancer and paracancerous tissues corresponding to patients with renal clear cell carcinoma who were treated at the Department of Urology,Subei People’s Hospital of Jiangsu Province and received surgical treatment from September 2019 to September 2020 were collected.Finally,use bioinformatics to analyze the potential molecular mechanism of GRK6 involved in the occurrence and development of kidney cancer and possible signal pathways.Results: The results of bioinformatics analysis showed that the expression level of GRK6 in cancer tissues was significantly higher than that in normal tissues.At the same time,the overall survival rate(OS)of the patients with high expression level of GRK6 was lower than that of patients with low expression level(HR=1.52,log-rank P=0.036).Immunohistochemical techniques showed that the positive rate of GRK6 expression in renal cancer tissues was higher than that in adjacent tissues,and the difference was statistically significant.Western blotting showed that the expression of GRK6 in clear cell renal cell carcinoma was higher than that in normal tissues,and this was a statistically significant difference.The expression level of GRK6 in clear cell renal cell carcinoma tissues has no statistically significant difference in patients of different ages,genders,tumor sizes,pathological grades,and clinical stages.Bioinformatics analysis found that in renal clear cell carcinoma,GRK6 participates in the molecular regulatory network including CCR8 and other genes,and the expression level of GRK6 is significantly positively correlated with CCR8.Conclusion: This study found that GRK6 gene expression levels in clear cell renal cell carcinoma tissues are elevated,and it is significantly associated with the prognosis of clear cell renal cell carcinoma patients.It plays an important role in the occurrence and development of renal clear cell carcinoma.It can be used as a potential biomarker for the diagnosis,treatment and prognosis of renal clear cell carcinoma.
