Study On Saposhnikoviae Radix Processing Technology And Quality Evaluation System Of Pieces

Objective: The quality of the collected Saposhnikoviae Radix medicinal materials was evaluated;the processing technology of Saposhnikoviae Radix prepared pieces was discussed,and its quality was evaluated in many aspects;comprehensive appearance and internal quality,the grades of Saposhnikoviae Radix prepared pieces were classified.Methods:1.24 batches of Saposhnikoviae Radix medicinal materials from 8 producing areas were collected and analyzed by LC-MS combined with chemometrics.2.The collected herbs were selected.Firstly,the differences of the head and root of the net preparation part of Saposhnikoviae Radix were discussed by using liquid mass spectrometry combined with multivariate statistical method.Secondly,the single factor investigation combined with response surface method was used to investigate the influencing factors such as slice thickness,drying temperature and drying time in the processing process of the slices,so as to optimize the optimal processing technology of the slices.3.Referring to the Chinese Pharmacopoeia,the Hong Kong Standard of traditional Chinese Medicinal Materials and related research reports,the quality evaluation system of Saposhnikoviae Radix prepared slices was studied combined with self-made slices and collected market slices.The identification,examination,extract,content determination and fingerprint of prepared slices were studied to evaluate the quality of Saposhnikoviae Radix slices.4.The diameter,color,extract and internal index components of 30 batches of Saposhnikoviae Radix prepared slices were determined,and the correlation analysis and principal component analysis were carried out with the help of statistical analysis software.cluster analysis was carried out on 30 batches of Saposhnikoviae Radix prepared slices,and the grade classification standard of Saposhnikoviae Radix prepared slices was established.Results:1.There are certain differences in Saposhnikoviae Radix medicinal materials from different producing areas,and there are obvious differences between Northeast China and other regions.According to the degree of contribution to the difference,26 different components were screened,and 7 of them were identified as: isoparsleyin B,5-O-methylvisamicoside,saponin,5-O-methylvisamiol,divaricataester A,cimicifuga,and3-O-angelica acetohydrol.2.In the comparative study of SDL and SR,it was found that SDL and SR can be clearly distinguished,showing a big difference.Through further statistical analysis,14 different compounds were obtained,and 6 of them were identified as: holmosol,5-O-methylvisamicoside,5-O-methylvisamiol,Cimicifugain,phelloptorin and divaricataester C.3.Response surface methodology shows that the best processing technology is slice thickness 3 mm and drying at 50 ℃ for 2.5 h.4.Based on the existing standards,the thin-layer identification and content determination methods of Saposhnikoviae Radix decoction pieces have been optimized,and the method for fingerprinting of Saposhnikoviae Radix decoction pieces has been established.The methodological inspection results are qualified,indicating that all methods used can identify or determine Saposhnikoviae Radix decoction pieces.In addition,the inspection items,extracts,and index component content results of Saposhnikoviae Radix Decoction Pieces all comply with the relevant regulations in the 2015 Chinese Pharmacopoeia.5.Among the 30 batches of Saposhnikoviae Radix decoction pieces studied,there is no first-class product;there are 14 batches of second-class product(FF-4～FF-6,FF-9,FF-16～FF-19,FF-25～FF-30).Among them,only the origin of FF-9 medicinal materials is Jilin,the origin of FF-19 medicinal materials is Hebei,and the origin of the other second-class Saposhnikoviae Radix decoction pieces is Inner Mongolia.The remaining 16 batches of decoction pieces are all stocks.The grading results have undergone independent sample t-test,and the quality of the graded samples has a significant difference(p<0.01), indicating that the grading basis is reasonable and reliable.At the same time,it indicates that the quality of Saposhnikoviae Radix decoction pieces whose medicinal material origin is Inner Mongolia is better.Conclusion: There are certain differences in different origins of Saposhnikoviae Radix;the processing technology of Saposhnikoviae Radix decoction pieces is reasonable and feasible;the quality evaluation system of Saposhnikoviae Radix decoction pieces is scientific and comprehensive;the classification basis of Saposhnikoviae Radix decoction pieces is reasonable,and the classification results show that the origin of Saposhnikoviae Radix decoction pieces is related to its quality.Provide scientific reference for quality evaluation of Saposhnikoviae Radix decoction pieces and medicinal material planting base.
