Abstract/Summary:
Ulcerative colitis(UC)is a chronic idiopathic autoimmune inflammatory disease.The pathogenesis and causes are not yet clear.The main lesions are limited to the large intestinal mucosa and submucosa,affecting the gastrointestinal system.Conventional treatments for UC include aminosalicylic acid drugs(Salazosulfapyridine,etc.),corticosteroid drugs(glucocorticoid,etc.),immunosuppressants(Azathioprine,cyclosporine A,etc.),biological agents,etc.,but these drugs can only provide relief and long-term use can cause serious side effects and drug dependence.Every year,many patients have recurrence of symptoms and eventually require surgical intervention.The development of more effective and safe new drugs has become an important demand for the treatment of UC.Human intestinal microbes participate in the regulation of the immune response in the intestine and affect metabolism.Studies have shown that the imbalance of the intestinal microbiota is closely related to the disease state of UC.Probiotics can inhibit the growth of pathogenic bacteria by blocking adhesion sites,competing for nutrients,increasing butyrate and short-chain fatty acids to inhibit the growth of pathogenic bacteria in the unbalanced intestinal flora,and can enhance the local immune response and improve the intestinal barrier function.Although there are reports on the treatment of UC by probiotics,their therapeutic effects are limited.It is necessary to tap the potential of their effects on diseases and develop more efficient microecological preparations.Traditional Chinese medicine(TCM)has a long history of treating bowel diseases and has formed a mature treatment system.Unlike the targeted therapy of western medicine,TCM has a better effect in maintaining homeostasis,preventing IVrecurrence,and relieving symptoms.Traditional Chinese Medicine Treatment UC is flexible in oral and external administration and has less adverse reactions.It is favored by patients who prefer "natural therapy" or have experienced the side effects of traditional medicines.However,this kind of treatment is slow to take effect and is not well-targeted,and cannot meet the treatment needs of many types of UC.This study aims to explore the therapeutic potential of compound intestinal microecological preparations combined with traditional Chinese medicine and probiotics on UC,verify the synergistic effect of Chinese medicines and probiotics,and the effect of compound intestinal probiotics on mesalazine(positive drug)Resistant effect of side effects.The research contents of this thesis are as follows:Take 45 male C57BL/6J mice aged 6-8 weeks and randomly divide them into 9groups(n = 5 mice/group),which are the blank group,the negative control group of dextran sodium sulfate(DSS)induction model,and the United States.Salazine positive control treatment group,compound probiotics treatment group(low,medium and high dose three treatment groups),mesalazine and compound probiotics compound treatment group,traditional Chinese medicine treatment group,probiotics treatment group.1.The therapeutic effect of compound intestinal probiotics on intestinal inflammation and injury of acute ulcerative colitis in mice induced by DSS.2.The effect of compound intestinal microecological preparation on the intestinal microbial ecology of mice with acute ulcerative colitis induced by DSS.The results show that the compound intestinal microecological preparation has a significant therapeutic effect on UC.It can repair the damaged colon tissue of DSS mice,reduce the infiltration of inflammatory factors,increase the percentage of beneficial bacteria and reduces the abundance of harmful bacteria,restore the richness and diversity of the intestinal flora.Mesalazine significantly reduced the damage of colon tissue in DSS mice,but it produced nephrotoxicity,reduced the abundance of harmful bacteria but failed to increase the abundance of beneficial bacteria,and affected the species diversity of intestinal flora.The therapeutic effect of the combined intestinal microecological preparation and mesalazine in the treatment group is better than that of each treatment group,the compound intestinal microecological preparation can restore the imbalance of intestinal flora caused by mesalazine and reduce renal interstitial ulcers.Traditional Chinese medicine does not show a significant therapeutic effect on short-term acute ulcerative colitis,but it can have a positive effect on the therapeutic effect of probiotics.In summary,the compound intestinal microecological preparation is more effective than mesalazine in restoring the imbalanced intestinal flora of DSS mice,and has a certain therapeutic effect on the side effects of mesalazine.Traditional Chinese medicine has a positive effect on probiotics,and its combination shows a better therapeutic effect than traditional Chinese medicine or probiotics.
