| ObjectiveOvarian cancer is one of the leading causes of death among female reproductive system tumors,and the latest cancer statistics show that it ranks fifth in the world in death rate.Epidemiology and genetic analysis showed that the occurrence and development of ovarian cancer is closely related to gene mutation,and its high mortality rate is caused by multiple factors,but metastasis and recurrence are the main reasons.Therefore,it is very important to search for molecular markers of poor prognosis of ovarian cancer and to study the mechanism of metastasis,and to inhibit the migration and invasion of tumor cells,so as to prolong the survival period of malignant tumor patients and improve their quality of life and prognosis.We found that laminin subunit α5(LAMA5)is highly expressed in ovarian cancer tissues,and the expression of LAMA5 is associated with survival and prognosis of ovarian cancer.In this study,the expression and function of LAMA5 in serous ovarian cancer cells were studied to explore its clinical significance.Methods1.The expression,clinicopathological features and prognosis of LAMA5 in serous ovarian cancer were analyzed by TCGA,GEO and other databases and immunohistochemistry,and the clinical significance of LAMA5 was evaluated;2.The effects of LAMA5 on proliferation,migration and invasion function of high-grade serous ovarian cancer cells were detected by cell CCK8,Transwell and other experiments;3.To predict the possible mechanism of LAMA5 in the development of ovarian cancer through GSEA.Results1.The results of database analysis and immunohistochemistry showed that the expression level of LAMA5 in epithelial ovarian cancer was significantly higher than that in normal ovarian tissue,and the prognosis of patients with high expression was poor;2.CCK8 and Transwell results showed that specific LAMA5 knockout significantly inhibited the proliferation,migration and invasion of high-grade serous ovarian cancer cells;3.LAMA5 may be associated with MITOTIC_SPINDLE and Hh signaling pathways in ovarian cancer by GSEA analysis.Conclusions1.LAMA5 is highly expressed in ovarian cancer tissues,and patients with high expression have poor prognosis;2.LAMA5 can promote the proliferation,migration and invasion of highgrade serous ovarian cancer cells,and has the potential to be a marker of poor prognosis of ovarian cancer. |
