Background:Ovarian cancer is one of the most common gynecological malignancies,the incidence only after endometrial cancer and cervical cancer.The current clinical treatment of ovarian cancer with surgery combined with radiotherapy and adjuvant chemotherapy based.Ovarian cancer patients,who Get the poor treatment,would accept immunotherapy,biological therapy and molecular targeted therapy and other methods of treatment.However,due to the large number of clinical manifestations of ovarian cancer,the onset of more hidden,spread quickly,poor efficacy.The incidence of younger and increasing every year,as well as the lack of effective early screening and diagnostic methods,the general diagnosis is already a late stage.Therefore,in the active search for more specific and sensitive early screening indicators and more effective treatment,we have to reduce the incidence of ovarian cancer,to minimize exposure to ovarian cancer-related factors.Method:Previous studies have shown that PFOA enhances the migration and invasion of breast cancer,colorectal cancer and endometrial carcinoma.To demonstrate whether PFOA has an effect on ovarian cancer,we have treated different concentrations of PFOA in human ovarian cancer cells A2780.The phenotype of ovarian cancer cells was observed by scratch experiment,CCK-8 and Transwell.The expression of ERK,p-ERK and MMP-2/-9 protein in the cells of different time were observed by using the concentration of PFOA in the ovarian cancer cells.The expression of ERK,p-ERK and MMP-2/-9 protein was detected by using MAPK inhibitor U0126 to block the signal pathway,as well as the expression of MMP-2/9 mRNA expression levels and changes in cell migration and invasion ability.Result:PFOA can enhance the migration and invasion of ovarian cancer cells,but no significant effect on proliferation.Drug concentration in the role of 100nM obvious,the effect of PFOA on the expression of MMP-2/-9 mRNA and protein expression in ovarian cancer cells was significantly higher than that in control group(P<0.05).The expression of MMP-9 protein increased at 3h and decreased slightly after 12h,the expression of MMP-2 protein increased from 3 h and increased with time.ERK phosphorylation began to increase at 3h,and began to weaken after 6h peak.In the medium,inhibitor U0126 was added respectively,It was found that U0126 could block ERK phosphorylation and MMP-2/-9 mRNA and protein expression,but also can reduce ovarian cancer cell migration and invasion ability.Conclusion:PFOA can enhance the migration and invasion of ovarian cancer cells and induce the expression of MMP-2/-9 by activating MAPK/ERK signaling pathway.
