Objective: To observe the changes of intestinal microflora in patients with psoriasis vulgaris with spleen deficiency and dampness-Sheng syndrome before and after oral administration of Spleen Detoxification Soup by 16 Sr DNA sequencing.The influence of raditional Chinese medicine on intestinal microflora of psoriasis was explored by comparing the experimental group with the healthy control group before and after treatment.Methods: The fecal samples of 20 patients with psoriasis vulgaris and 20 healthy controls with spleen deficiency and dampness were collected and analyzed by 16 Sr DNA.Results: 1.The gender ratio of the 20 included patients was 1.86:1,with an average age of43.9 years.By comparing the severity of various parts of the body,it can be found that the trunk and lower limbs are different from the head,and the scales of each part are more obvious than those of other parts.There is no significant difference in erythema and infiltration of each part of the body.2.The sample after quality control,with 97% of the consistency will become OTUs sequence clustering,received 3645 OTUs,then the OTUs sequence annotation with Silva138 database for species.In the annotation results,a total of 1,589(43.59%)OTUs were annotated to the genus level.At the phylum level,we found that the dominant ones were Firmicutes,Proteobacteria,and Actinobacteriota.At class level,the dominant ones were Clostridia,Gammaproteobacteria and Actinobacteria.At order level,the dominant species were Enterobacterales,Lachnospirales,and Bifidobacteriales.At the family level,the dominant species were Enterobacteriaceae,Lachnospiraceae,and Bifidobacteriaceae.At the genus level,the dominant species were Escherichia-Shigella,Bifidobacterium and Bacteroides.The dominant species were Escherichia_coli,Bacteroides_Vulgatus and Ruminococcus.3.Alphadiversity can determine whether there are significant differences in the overall community structure between different populations.Conventional T-test showed that Bacteroidota and Bifidobacteriales were significant between the experimental group and the experimental group before and after treatment(P < 0.05),and there were differences between the healthy control group and the experimental group.Through the LEFSE analysis of A-B-C,there were 11 LDAScore > 4 biomarkers,that is,there were statistically significant differences in the abundance between groups.Betadiversity used PCo A and PCA to analyze the differences between groups,and it was found that there was a significant difference between the distance before and after treatment between the healthy control group and the experimental group.Conclusion: This study found that the structure of intestinal flora of patients with psoriasis was different from that of healthy control group.The intestinal flora of experimental group was different before and after oral SDS.This paper provides a new method to explore the effect of SDS on the regulation of intestinal microorganism in patients with psoriasis.
