| Objective:To observe the effect of Chai Shao Ru Pi Pellet on the expressions of MVD and VEGF proteins in rats breast precancerous lesion model,and explore the intervention mechanism of Chai Shao Ru Pi Pellet in the prevention and treatment of precancerous breast cancer by animal experiments.Methods: Sixty rats were randomly selected as control group,10 were randomly selected as blank control group.The remaining 50 were for the model preparation group.DMBA(7,12-Dimethyl benzanthracene)was used as an initial gavage for modeling.After 9 weeks of routine feeding,1 and 5rats were randomly selected from the blank control group and the model preparation group respectively,and sacrificed.The mammary tissue of the second and third rats was examined by pathology.After confirming the success of modeling,the remaining 45 rats in the model preparation group were randomly divided into model group,Chinese medicine(low/medium/high)dose group and positive control group,a total of 5groups with 9 rats in each group.The blank control group and model group were given normal saline(10ml/1kg/d)by gavage,the traditional Chinese medicine(low/medium/high)dose group was given Chai Shao Ru Pi Pellet(0.32/0.64/1.28g/1kg/d)by gavage,and the positive control group was given Tamoxifen Citrate Tablets(13.2mg/kg/d)by gavage.The intervention time was 15 days.The second day after the intervention,all rats were sacrificed,and the second and third breast tissues of the rats were resected.HE staining was used for breast histomorphology detection,and SP immunohistochemical method was used to determine the expression of MVD and VEGF protein in the breast tissues of the rats.SPSS22.0 software was used for statistical analysis.Results:(1)Pathomorphology:Intervention of medium and high doses of Chai Shao Ru Pi Pellet and Tamoxifen Citrate Tablets in rats breast precancerous lesion model could improve the pathomorphological distribution of mammary gland tissue without statistical significance,while low doses of Chai Shao Ru Pi Pellet had no significant effect on the improvement of pathomorphology.(2)Immunohistochemistry:High-dose Chai Shao Ru Pi Pellet and Tamoxifen Citrate Tablets could reduce the expression of MVD in the mammary tissue of model rats,and there was no statistical difference,while low-dose and medium-dose could not significantly improve the expression of MVD;The positive expression intensity of VEGF in medium and high doses of Chai Shao Ru Pi Pellet and Tamoxifen Citrate Tablets decreased compared with before,and there was no statistical difference between the medium and high dose groups;These results indicated that there were dose-effect differences in the inhibition of MVD and VEGF by Chai Shao Ru Pi Pellet.Conclusions:The results suggest that DMBA initial one-time gavage method can establish rats models of breast precancerous lesion.Chai Shao Ru Pi Pellet can reduce MVD of breast cancer precancerous tissue,and reduce the expression of VEGF protein in the tissue,and have the effect of reducing the strength of microvascular formation and angiogenesis.Chai Shao Ru Pi Pellet has a certain function in the prevention and treatment of breast cancer precancerous lesions. |
