ObjectiveTo observe the effects at “Fei shu”,“Da zhui” and “Feng men” points on the expression of FIZZ1 and NOTCH1 in lung tissue of asthmatic model rats in each group,and to explore the mechanism of “Shao’s Method of Wu Zhen” on airway remodeling of asthma,so as to provide a more scientific t heoretical basis for the mechanism study of acupuncture and moxibustion in the treatment of asthma.MethodFort y male SPF SD healthy rats were randomly divided into blank group,model group,acupuncture group and moxibustion group,with 10 rats in each group.The first and eighth days of the experiment were sensitized by intraperitoneal injection of 10% compound OVA antigen solution 1 ml.At the fifteenth day,the asthmatic rat model was established by inhaling 1% OVA solution for 20 minutes(once a day,7 times in total).The rats in the treatment group were all selected “Fei shu”,“Da zhui” and “Feng men”.After treatment Acupuncture group and moxibustion group were given provocation.The blank group was stimulated by intraperitoneal injection and aerosol inhalation with saline.The rats were anesthetized with 10% chloral hydrate after the last treatment.The thoracic cavity was exposed.The right middle and lower lobe lung tissues(including hilum)were extracted and fixed in 4% paraformaldehyde for one week.Pathological sections were prepared by paraffin embedding,hematoxylin-eosin(HE)staining and immune histochemical staining of lung tissue.The morphological changes of lung tissue and the expression of FIZZ1,NOTCH1 and α-SMA in lung tissue of rats in each group were observed,and the correlation between FIZZ1,NOTCH1 and α-SMA was analyzed.Results1 The behavior of rats of each groupCompared with the blank group,the rats in the model group had shortness of breath,wheezing,scratching ears and cheeks,nervous tension and slow reaction.The rats in the blank group breathed evenl y and had stable mood without abnormal mani festations.Compared with the model group,the symptoms of the acupuncture treatm ent group and the moxibustion treatment group were lighter,more sensitive and quiet er.2 Lung tissue morphology of each groupIn the blank group,the airway tissue structure was intact without inflammatory cell infiltration.In the model group,the pulmonary tissue structure was disordered,the bronchial lumen was narrow,the wall was thickened,the mucus secreted in the lumen was more,the epithelial cells fell off,a large number of inflammatory cells appeared near the blood vessels and bronchi,the alveolar wall was broken and collapsed,and the smooth muscle was thickened.The airway structure of the acupuncture treatment group and the moxibustion treatment group was basically intact,the endocrine mucus of the bronchus was less,and a small amount of inflammatory cells appeared.3 Comparison of FIZZ1 of each groupThe expression of FIZZ1 in model group was significantl y higher than that of control group(P<0.05),and the positive expression of FIZZ1 in two treatment groups were significantl y lower than that of model group,but still higher than t hat of blank group(P<0.05),and the expression of FIZZ1 in lung tissue of acupuncture group was less than that of moxibustion group,the difference was significant(P<0.05).There was statistical significance(P<0.05).4 Comparison of NOTCH1 of each groupCompared with the model group,the expression of NOTCH1 in the two treatment groups were significantl y lower than that in the model group(P<0.05),and the difference was statistically significant(P<0.05).Compared with the blank group,the expression of NOTCH1 in the two treatment groups were significantly lower than that in the model group(P<0.05).The positive expression of FIZZ1 in moxibustion group was higher than that in moxibustion group(P<0.05).5 Comparison of NOTCH1 of each groupCompared with the blank group,a large number of α-SMA positive cells appeared in the lung tissue of the model control group rats,the difference between the two groups was statistically significant(P<0.05);the content of α-SMA in the lung tissue of the treatment groups rats were higher than that of the blank control group,but significantl y lower than that of the model control group,the difference was statistically significant(P<0.05).Compared with the moxibustion treatment group,there was no significant difference between two treatment groups(P> 0.05).6 Relevance ansl ysisFIZZ1 and NOTCH1 had strong correlation with α-SMA.The correlation coefficients r were 0.899 and 0.889,with statistical significance(P<0.01).7 Anal ysis of Acupuncture and Moxi bustionAcupuncture and moxibustion can regulate the contents of FIZZ1,NOTCH1 and α-SMA,especially FIZZ1 and NOTCH1,which can reduce the proliferation of smooth muscle and inhibit airway inflammation and airway remodeling.Conclusion1 “Shao’s Method of Wu Zhen” can effectively improve the symptoms and behavioral signs of asthmatic model rats.2 “Shao’s Method of Wu Zhen” can effectively improve the morphological changes of lung tissue in asthmatic model rats.Airway inflammation and airway remodeling can be improved during the occurrence and development of asthma.3 “Shao’s Method of Wu Zhen” can reduce the expression of FIZZ1,NOTCH1 and α-SMA.4 FIZZ1 and NOTCH1 are closely related to α-SMA,which can induce the differentiation of fibrobl asts and play a s ynergistic role in promoting airway remodeling in asthma.5 “Shao’s Method of Wu Zhen” can inhibit the inflammatory infiltration of lung tissue and the destruction of lung structure in asthmatic model rats,reduce smooth muscle proliferation,and improve the pathological link of airway inflammation and airway remodeling during the occurrence and development of asthma.
