| Background and Objective: With the rapid development of China’s national economy,the significant improvement of the people’s material living standards and the continuous extension of the population’s life span,the proportion of cardio-cerebrovascular disease mortality is also increasing.Coronary artery disease(CAD)and ischemic stroke(IS)have become a huge challenge facing humanity today.Although CAD and IS belong to two different vascular systems,their risk factors,pathophysiology,and prevention and treatment overlap.Many studies have shown that certain gene changes can affect blood lipid metabolism,but also affect cardio-cerebrovascular diseases.Some data indicate that certain axonemal heavy chain dynein type 11 gene(DNAH11)single nucleotide polymorphisms(SNPs)are associated with blood lipid levels and ischemic cardio-cerebrovascular disease,but the results are also inconsistent.Therefore,the purpose of this study was to explore the association between the DNAH11 rs12670130 SNP and serum lipid levels and the risk of CAD and IS in the Guangxi Han population.Methods: The Han residents living in Guangxi Zhuang Autonomous Region were selected as the research object and divided into a control group and a case group.Among them,619 subjects in normal control group,560 cases in CAD group and 543 cases in IS group.The general clinical data of the research subjects were collected in the form of questionnaires and physical examinations,and venous blood was taken for serum lipid determination,DNA extraction and genotype determination.The genotypes of the DNAH11 rs12670130 SNP in the samples were obtained by the Snapshot technology.Results: 1.There were statistically significant differences in serum low-density lipoprotein cholesterol(LDL-C)levels among different genotypes of the DNAH11 rs12670130 SNP in the normal control group(P < 0.05).The carriers of the rs12670130 G allele had higher LDL-C levels than the non-carriers of the rs12670130 G allele.2.DNAH11 rs12670130 SNP is not significantly associated with the risk of CAD.AA vs.GA: OR = 1.036,95% CI= 0.791-1.358,P > 0.05;AA vs.GG: OR = 0.623,95% CI = 0.319-1.215,P >0.05.3.There was no significant association between the DNAH11 rs12670130 SNP and the risk of IS.AA vs.GA: OR = 0.958,95% CI = 0.732-1.253,P >0.05;AA vs.GG: OR = 0.788,95% CI = 0.416-1.490,P > 0.05.4.Stratified analysis showed that the rs12670130 SNP was not associated with the risk of CAD and IS according to the risk factors such as gender,age,BMI,smoking and drinking(P > 0.05 for all).Conclusions: The DNAH11 rs12670130 SNP was significantly correlated with serum LDL-C levels,the carriers of rs12670130 G allele had higher serum LDL-C levels than the subjects with rs12670130 AA homozygotes,but the DNAH11 rs12670130 SNP was not associated with the risk of CAD and IS.It is suggests that the DNAH11 rs12670130 SNP does not affect the incidence of CAD and IS in Guangxi Han population through blood lipid metabolism. |
