| Background: Preeclampsia(PE)is a kind of hypertension disease in pregnancy,with threating to the health of pregnant women and fetus.There are many theories about its etiology and pathogenesis.Inflammatory immune overactivation of the maternal fetal interface and the whole body is one of them.Placental protein 14 is an immunomodulator related to pregnancy closely.Objective:Through comparing the different of placental protein 14 and Th17,Treg related cytokines IL-17,IL-10 in decidual tissues of normal pregnant women and preeclampsia patients,and the expression of placental protein 14 and Th17,Treg related transcription factors RORC,Foxp3,to investigate the role and relationship of placental protein 14 and Th17,Treg in the pathogenesis of preeclampsia.Methods: Selecting case-control study,15 cases of preeclampsia were randomly selected as study group and 15 cases of normal pregnancy at the same period as control group.ELISA was used to detect the levels of placental protein14,cytokines IL-17 and IL-10,and the real-time quantitative PCR was used to detect the expression of PP14 m RNA,FOXP3 m RNA and RORC m RNA in decidual tissues of two groups.Analyzing and comparing the expression differences between two groups through spss20.0 software.Result: The expression of PP14 m RNA in decidua of preeclampsia patients was lower than that in normal control group,and the difference was statistically significant(P < 0.05).The expression level of FOXP3 m RNA was lower than that of the normal control group,and the difference was statistically significant(P < 0.05).The expression of RORC m RNA was higher than that of normal control group,and the difference was statistically significant(P < 0.05).The concentration of PP14 in decidual tissue of preeclampsia was lower than that of normal control group,with statistically significant difference(P < 0.05).The concentration of IL-10 in the experimental group was significantly lower than that in the normal control group,and the difference was statistically significant(P < 0.05).The concentration of IL-17 in the experimental group was significantly higher than that in the normal control group,the difference was statistically significant(P < 0.05).PP14 was positively correlated with IL-10 and FOXP3(P < 0.05).Conclusion: The decrease of Treg cells in decidua of preeclampsia patients was related to the decrease of PP14,and the imbalance of Th17/Treg cells in the maternal-fetal interface of preeclampsia may be related to the decrease of placental protein 14. |
