| Objective : According to the proportion of Th17 cells and Treg cells in CD4+ T lymphocytes in decidual tissues of missed abortion patients and normal early pregnant women,combined with the difference in expression of related cytokines IL-17,IL-21,IL-10 and TGF-? in decidual tissues of the two groups,explore the role of Th17/Treg immune imbalance in the occurrence and development of missed abortion.Methods : 30 missed abortion patients in the first affiliated hospital of guangxi medical university from September 2017 to June 2018 were selected as the experimental group,and 30 normal early pregnancy patients who required abortion during the same period were selected as the control group.The diagnosis was based on transvaginal ultrasonography and the dynamic monitoring results of human chorionic gonadotropin(HCG)levels.Flow cytometry(FCM)was used to detect the proportion of Th17 cells and Treg cells in CD4+T lymphocytes in decidua tissues of the two groups respectively,and the expressions of IL-17,IL-21,IL-10 and TGF-? in decidua tissues of the two groups were detected by immunohistochemistry(IHC).SPSS 22.0 statistical software was used to calculate the above measurement data by t test withcompletely random design for comparison between the two samples,and the differences between the two groups were analyzed and compared.Result : The percentages of Th17 cells in the experiment group and the control group were(7.21±1.30)and(5.91±1.06),the percentages of Treg cells in the experiment group and the control group were(5.75±1.05)and(7.38±1.25),the ratio of Th17/Treg in the experiment group and the control group were(1.31±0.41)and(0.82±0.18),Compared with the two groups,P < 0.05,the difference were statistically significant.The average optical density(OD)of IL-17 in the experiment group and the control group were(0.058±0.007)and(0.047±0.009),the OD of IL-21 in the experiment group and the control group were(0.030±0.009)and(0.024±0.006),the OD of IL-10 in the experiment group and the control group were(0.028±0.010)and(0.035±0.009),and the OD of TGF-? in the experiment group and the control group were(0.026±0.007)and(0.038±0.008),P<0.05,the difference was statistically significant.Conclusion : The percentage of Th17 cells in CD4+T lymphocytes and the ratio of Th17 to Treg and the expression of Th17-related cytokines IL-17 and IL-21 in decidua tissues of the experiment group patients were increased,while the percentage of Treg cells in CD4+T lymphocytes and the expression of Treg-related cytokines IL-10 and TGF-? were decreased,suggesting Th17/Treg immune imbalance was probably involved in the occurrence and development of missed abortion. |
