adjacent tissues and its relationship with HCC clinical data,we explored whether there is a difference in the expression of miRNA-182 in liver cancer and normal tissue.Transfection results in high and low expression of miRNA-182.Through in vitro cell function experiments,the biological function of miRNA-182 in hepatoma cell SMMC-7721 and its potential mechanism are studied to provide a basis for finding new biological targets for the treatment of hepatocellular carcinoma.Methods:1.Collect the surgical specimens of patients diagnosed with liver cancer in the clinic,divide them into cancer tissue group and adjacent cancer tissue group,use the Trizol method to extract the total RNA of the tissue,perform qRT-PCR detection after reverse transcription,statistical analysis The expression differences in 73 pairs of HCC cancer tissues and adjacent tissues,and the relationship between their expression levels and the clinical data of HCC patients,were calculated using chi-square test and measurement data using T test.2.In vitro culture of hepatoma cells SMMC-7721,using gene transfection technology,using miRNA-182 mimic mimic and inhibitor inhibitor to transfect SMMC-7721 hepatoma cells to transfect miRNA-182 mimic and miRNA-182 inhibitor Liver cancer cells were used as the experimental group,and liver cells of mimic NC and inhibitor NC that had been transfected were used as the control group.The effect of high and low expression of miRNA-182 on the proliferation ability of SMMC-7721 hepatocellular carcinoma cells was verified by the MTT cell proliferation experiment;the horizontal and vertical migration ability of miRNA-182 was verified by cell scratch repair experiment and Transwell migration experiment Impact;Transwell invasion test was used to detect the change of invasion ability after transfection with miRNA-182.Result:1.The expression analysis of miRNA-182 in hepatocellular carcinoma tissues and adjacent tissues showed that the expression level of miRNA-182 in hepatocellular carcinoma tissues was significantly lower than that in adjacent tissues,statistical significance(P<0.05).2.Analysis of the expression level of miRNA-182 and the clinicopathological characteristics of hepatocellular carcinoma showed that the expression level of miRNA-182 was related to the level of alpha-fetoprotein(AFP)(P <0.05).3.After transfection of miRNA-182 mimic mimic and inhibitor inhibitor,the expression level of miRNA-182 in SMMC-7721 hepatoma cells of miRNA-182 mimic is significantly higher than that of miRNA-182 in control group mimic NC,statistical significance(P<0.05).The expression level of miRNA-182 in SMMC-7721 hepatoma cells with miRNA-182 inhibitor after transfection was lower than that in SMMC-7721 hepatoma cells with miRNA-182 inhibitor NC after transfection,statistical significance(P<0.05).4.MTT cell proliferation experiments showed that the cell activity of SMMC-7721 liver cells transfected with miRNA-182 mimic was higher than that of SMMC-7721 liver cells transfected with mimic NC,statistical significance(P<0.05).The cell activity of miR-182 inhibitor hepatocellular carcinoma SMMC-7721 is lower than that of SMNC-7721 transfected with inhibitor NC,statistical significance(P<0.05).5.The results of cell scratch repair experiment and Transwell migration experiment showed that compared with SMMC-7721 liver cancer cell transfected with mimic NC,SMMC-7721 liver cancer cell transfected with miRNA-182 mimic had poor cell repair ability and cell migration.The number is small,statistical significance(P=0.045;P=0.001);the cell repair ability and cell migration number of the hepatocellular carcinoma SMMC-7721 transfected with miR-182 inhibitor is smaller than that of the hepatocellular carcinoma SMMC-7721 transfected with inhibitor NC High,statistical significance(P=0.045;P<0.000).6.The results of Transwell invasion experiments showed that the number of cells transfected with miRNA-182 mimic hepatoma cells SMMC-7721 passed through the chamber was less than the number of cells transfected with miRNA-182 mimic hepatoma cells SMMC-7721,statistical significance(P<0.000);the number of cells transfected with SMR-7182 transfected with miR-182 inhibitor was greater than that of SMMC-7721 transfected with inhibitor NC,statistical significance(P<0.000).Conclusion:The expression level of miR-182 in liver cancer tissues is significantly higher than that in adjacent tissues,which may play a role in promoying the development of cancer in the occurrence and development of tumors.Up-regulating the expression of miR-182 can enhance the proliferation,migration and invasion ability of hepatoma cell line SMMC-7721.Mi R-182 is expected to become a new tumor marker and therapeutic target for liver
