Objective:To establish a method for the identification of Swertia L.by Fourier transform infrared(FTIR)spectroscopy combined with stoichiometry chemometrics methods.Compared with Swertia leducii,the content of the main active components of Swertia cincta was analyzed and the chemical fingerprint was established.The anti-fibrosis effect and its mechanism was studied,so as to compare the isotropy and difference of its chemical components and pharmacodynamics,and also provide a basis for the expansion of resources.Method:(1)FTIR spectroscopy and chemometrics were used to analyze the phylogenetic relationship of Swertia leducii and its related species.(2)The main active substances of Swertia leducii and Swertia cincta were analyzed and determined by HPLC and chemical fingerprints were established.(3)The MTT method was used to detect the inhibitory effect of alcohol extracts of Swertia leducii and Swertia cincta on rat hepatic stellate cells(HSC-T6).(4)CCL4 was used to establish the chemical liver fibrosis model.Three doses(0.2 g/kg,0.4 g/kg,0.8 g/kg)of the extract of Swertia leducii and Swertia cincta were administered by intragastric administration.,1 times a day for eight weeks.The concentrations of ALT,AST,total protein(TP),albumin(Alb)and total bilirubin(TBIL)were detected,and the morphological characteristics of liver pathological tissue sections were observed,etc.Results:(1)The main absorption area of Swertia leducii and and its related species was around 900~400 cm-1,1310~900 cm-1,1500~1310 cm-1,1800~1500 cm-1,2800~3000 cm-1,3000~3500 cm-1.The absorption peaks of the second derivative spectra were significantly different in the 400-1000 cm-1 fingerprint region,and the differences in peak number,peak strength and peak pattern were significant among the species Swertia cincta and Swertia nervosa had closed relationship with Swertia leducii(2)The common pattern of fingerprints and similarity evaluation was established of Swertia leducii and Swertia cincta.The results showed that two species have similar chemical components,but the contents of the three main active components were significantly different(3)In vitro anti-liver injury experiments showed that the inhibitory effects of Swertia leducii and Swertia cincta on the proliferation of HSC-T6 cells increased with the increasing concentration of the drug,indicating that the extracts had anti-liver fibrosis effects(4)The results of anti-liver injury experiments in vivo showed that the contents of ALT,AST,TBIL,TP and Alb in serum liver function indexes were significantly decreased(P<0.05),indicating that Swertia leducii and Swertia cincta had similar anti-liver fibrosis effects,and their effects were similar.Conclusion:FTIR spectroscopy combined with chemometrics methods can distinguish different species of genus Swertia L.and display the genetic relationship between Swertia leducii and its relative.According to the comparative study of the main active components and the anti-liver injury effect of Swertia leducii and Swertia cincta,the results showed that Swertia cincta had potential as a new drug source,and its anti-liver injury effect deserve deeply researching. |
