| Objective: To detect the expression level of histone methyltransferase Ezh2 in peripheral blood mononuclear cells and the levels of IL-6 and IL-17 in peripheral blood of patients with primary immune thrombocytopenia(ITP),analyze the differences of Ezh2,IL-6 and IL-17 levels between ITP patients and normal people,and the differences of Ezh2 expression levels in different TCM syndromes,and analyze the differences between Ezh2 and IL-6,IL-17 levels.The correlation between platelet count and Ezh2,IL-6,IL-17 was discussed.The relationship between Ezh2 gene and ITP was discussed,which provided new ideas for the diagnosis and treatment of ITP.Methods: Thirty-two patients with ITP were collected and classified according to TCM syndromes: 9 cases of blood-heat delusion syndrome,11 cases of Yin-Deficiency and blood-heat syndrome,12 cases of Qi-deficiency deficiency syndrome.At the same time,peripheral blood of 12 normal persons were collected as control group.The expression of Ezh2 in peripheral blood mononuclear cells was detected by real-time fluorescence quantitative PCR,the levels of IL-6 and IL-17 in peripheral blood were measured by Elisa method,and the expression levels of Ezh2 and IL-6,I in ITP patients were analyzed.The level of L-17 was different from that of normal people.The expression of Ezh2 in different syndromes was analyzed.The correlation between Ezh2 and IL-6,IL-17,platelet count and expression of Ezh2,IL-17 and IL-6 were studied.Results:1.The expression level of Ezh2 m RNA in ITP patients was significantly higher than that in control group(P < 0.01).The levels of IL-6 and IL-17 in ITP patients were significantly higher than those in control group(P < 0.01);2.The expression level of Ezh2 m RNA in ITP patients was positively correlated with that of IL-6(R = 0.912,P <0.01).The expression level of Ezh2 m RNA in ITP patients was positively correlated with that of IL-17(R = 0.893,P < 0.01);3.The platelet count in ITP patients was negatively correlated with Ezh2 gene expression level(R=-0.724,P<0.01),significantly negatively correlated with IL-6 level(R=-0.809,P<0.01)and negatively correlated with IL-17 level(R=-0.885,P<0.01);4.Compared with the normal control group,the expression level of Ezh2 gene in patients with blood-heat delusion syndrome was P=0.092 > 0.05,with no statistical significance.The level of Ezh2 gene expression in patients with deficiency of Qi was higher than that in normal control group(P = 0.013 <0.05).There was significant difference between different TCM syndromes groups(P >0.05).Conclusion:The expression level of Ezh2 m RNA in ITP patients was higher than that in control group,and the levels of IL-6 and IL-17 in ITP patients were higher than that in control group.The expression of Ezh2 m RNA was positively correlated with the levels of IL-6 and IL-17,suggesting that Ezh2 may affect the secretion of cytokines through the regulation of immune system and participate in the pathogenesis or disease process of ITP.The expression level of Ezh2 m RNA in patients with Yin deficiency,blood-heat syndrome and Qi deficiency deficiency deficiency was higher than that in normal control group,which had statistical significance.There was no statistical significance between the expression level of Ezh2 m RNA in patients with blood-heat delusion syndrome and that in normal control group.There was no significant difference among different TCM syndromes.The expression level of Ezh2 might not be suitable as a reference for microcosmic syndrome differentiation index of ITP TCM syndromes. |
