Objective: For patients with polycystic ovary syndrome who were treated with in vitro fertilization-embryo transfer,the patients with polycystic ovary syndrome were treated with Bushen Chinese medicine to observe the clinical effect of improving the egg cell quality of PCOS patients with kidney qi deficiency syndrome,and to detect the expression of PI3K/FOXO3 a protein and m RNA in ovarian granulosa cells.Change to explore its mechanism of action.Methods: Sixty patients with renal qi-deficiency syndrome who received IVF-ET were randomly divided into observation group and control group,30 cases in each group.30 patients with IVF-ET were selected as blank group.All three groups of patients used the superovulation long program.The observation group received 1 bag po TID from the second day of menstruation on the third day of menstruation from the second day of menstruation,and the treatment was performed for 3 menstrual cycles.The control group and the blank group were not treated with traditional Chinese medicine.The follicular fluid was collected on the day of egg retrieval,and the granulosa cells were cryopreserved.The expression of PI3K/FOXO3 a m RNA and protein was determined by RT-PCR and Western blotting.The three groups of patients were enrolled in the group and HCG Japanese kidney deficiency syndrome.The change of syndrome scores was evaluated;the number of eggs obtained in three groups,fertilization rate,embryogenesis rate,and embryo rate were evaluated.Results: 1.The observation group’s kidney qi deficiency syndrome TCM syndrome quantitative score decreased after treatment compared with before treatment,the results were statistically significant(P<0.01),kidney qi deficiency symptoms were improved.2.Compared with the control group,the observation group had a low dose of Gn and a small number of days,and the difference was statistically significant(P<0.05).3.There were significant differences in fertilization rates among the three groups.Among them,the observation group had the highest fertilization rate,the blank group was the second,and the control group was the lowest.There was significant difference between the observation group and the control group(P<0.01).There was significant difference between the blank group and the control group(P<0.05).There was no statistical difference between the observation group and the blank group.Academic significance(P>0.05).4.The rates of transplantable embryos in the three groups were different,and the rate of transplantable embryos in the observation group and the blank group was significantly higher than that in the control group.The difference was statistically significant(P<0.01).The observation group was compared with the blank group(P>0.05),no significant statistical difference.5.There was no significant difference in the quality of embryos between the three groups(P>0.05).There was no significant difference between the two groups(P>0.05).6.The expression of PI3 K m RNA in the control group was higher than that in the observation group and the blank group.The expression of FOXO3 a m RNA in the observation group was higher than that in the control group and the blank group.However,there was no significant difference in PI3 K m RNA and FOXO3 a m RNA expression between the three groups(P>0.05).7.The PI3 K protein expression was significantly different between the three groups(P<0.01).The control group had the highest PI3 K protein expression,andthe difference was statistically significant compared with the observation group and the blank group(P<0.05).The observation group and the blank group There was no significant difference in PI3 K protein expression between the two groups(P>0.05).8.The expression of FOXO3 a protein in the observation group showed an increasing trend,but the difference between the three groups was not statistically significant(P>0.05).Conclusion: Erzhi Tiaojing Granule can can effectively improve the symptoms of kidney qi deficiency in patients with kidney qi deficiency syndrome PCOS,and increase the fertilization rate and the rate of transplantable embryos.The mechanism may be related to the down-regulation of PI3 K expression and up-regulation of FOXO3 a in granulosa cells.
