Purpose:To study the anti-atherosclerosis mechanism of Jiawei Simiao Yongan Tang granules by regulating PI3K/Akt signaling pathway,we detect the expression of LC3,Beclin 1 protein and m RNA and Akt,mTOR proteins regulated by PI3K/Akt signaling pathway in aortic arch tissue of atherosclerotic rabbits.Liver function,blood lipid and hs-CRP were measured for clarifying the role of PI3K/Akt signaling pathway in the occurrence and development of atherosclerosis.In order to explore the specific mechanism of anti-inflammatory and anti-atherosclerosis of Jiawei Simiao Yongan Tang granules,the PI3K/Akt signaling pathway in aortic arch tissue of atherosclerotic rabbits was further interfered with by Jiawei Simiao Yongan Tang granules and to provide theoretical basis for clinical use of Jiawei Simiao Yongan Tang granules,and to provide traditional Chinese medicine treatment for atherosclerosis.Methods:Experiment 1: Sixteen male Japanese big ear white rabbits were randomly divided into normal group and model group.Eight rabbits in each group were killed after 8 weeks of high fat diet.The general condition,liver function,blood lipids,hs-CRP,pathological changes of aortic arch and the expression of LC3,Beclin 1 protein and m RNA and Akt,mTOR proteins were observed in the two groups.Experiment 2: The second experiment was conducted under the condition that the difference of autophagy level between the two groups was statistically significant.48normal-grade male Japanese big-eared white rabbits were randomly divided into normal group,model group,Ale group,low-dose group of Jiawei Simiao Yongan Tang granules,medium-dose group of Jiawei Simiao Yongan Tang granules and high-dose group of Jiawei Simiao Yongan Tang granules,with 8 rabbits in each group.High-fat diet was used to make the model.After the model was successfully established,the rabbits in six groups were given distilled water and distilled water 10ml/kg,atorvastatincalcium 0.045mg/kg,Jiawei Simiao Yongan Tang granules 1.8g/kg,3.6g/kg and7.2g/kg suspension for 8 weeks.The animals were killed at the end of the 8th week.Detection index is the same as experiment 1.Results:Experiment 1 :1.Liver function,blood lipid and hs-CRP index: In normal group and model group,liver function was normal,and high density lipoprotein cholesterol(HDL-Ch)had no significant difference(P > 0.05).Compared with normal group,total cholesterol(TCh),triglyceride(TG),low density lipoprotein cholesterol(LDL-Ch),and high sensitivity C-reactive protein(hs-CRP)in model group increased significantly(P< 0.05).2.HE staining of aortic arch: Compared with the normal group,the rabbit model of the aortic arch of the model group was thickened and the surface was not smooth,and there were a large number of plaques.3.PI3K/Akt signaling pathway: Compared with the normal group,the expression of p-Akt and p-mTOR protein in the model group was increased,the expression of Akt and mTOR protein was not significantly different,and the expression of Beclin 1 and LC3 protein and m RNA was decreased,the difference was statistically significant(P < 0.05).Experiment 2 :1.Liver function,blood lipid and hs-CRP index: The liver function of each group was normal,and there was no significant change in high-density lipoprotein cholesterol(HDL).Compared with the model group,the total cholesterol(TCh),triglyceride(TG),low-density lipoprotein cholesterol(LDL-Ch)and high-sensitivity C-reactive protein(hs-CRP)of the high-dose group and the Ale group after treatment were decreased,with statistical difference.Meaning: There was no significant difference between the low and middle dose groups of Jiawei Simiao Yongan Tang granules(P > 0.05).2.HE staining of aortic arch: Compared with the model group,the rabbits in the low-dose and middle-dose groups of Jiawei Simiao Yongan Tang had thickened intima of the aortic arch,the surface was not smooth,and there were scattered plaques;Therabbit intima of the aortic arch in the Ale and he high-dose groups of Jiawei Simiao Yongan Tang granules was smooth and had a small amount of plaque.3.PI3K/Akt signaling pathway: Compared with the model group,the expression of p-Akt and p-mTOR protein in Ale group and high dose group of Jiawei Simiao Yongan Tang granules was decreased,the expression of Beclin 1,LC3 protein and m RNA was increased,the difference was statistically significant(P < 0.05);while there was no significant difference between Ale group and high dose group of Jiawei Simiao Yongan Tang granules(P > 0.05).There was no significant difference in the expression of Akt,mTOR,Beclin 1 and LC3 between the low and medium dose groups of Jiawei Simiao Yongan Tang granules(P > 0.05).Conclusions:1.In the aortic arch tissues of atherosclerotic rabbits,the expression of p-Akt and p-mTOR protein was increased,but the expression of Akt and mTOR protein did not differ significantly,while the expression of Beclin 1,LC3 protein and m RNA was decreased.2.Jiawei Simiao Yongan Tang granules has the effect of improving atherosclerosis.3.The mechanism of Jiawei Simiao Yongan Tang Granules on improving atherosclerosis may be through regulating PI3K/Akt signaling pathway,that is,by inhibiting Akt and mTOR signaling pathway,the expression of p-Akt and p-mTOR protein is reduced,while the expression of Beclin 1,LC3 protein and m RNA is increased,thereby enhancing the autophagy of aortic arch tissue cells and exerting anti-atherosclerosis effect.
