| Ajuga ovalifolia var.calantha belongs to genus Ajuga of family Labiatae is the dry grass of Ajuga ovalifolia Bur.et Franch.var.calantha(Diels ex Limpricht)C.Y.Wu et C.Chen.This dissertation investigated on chemical constituents of the extract of Ajuga ovalifolia var.calantha.The purpose is to enrich the Ajuga ovalifolia,and to screen the anti-inflammatory and antitumor constituents with high efficiency and low toxicity,laying the foundation for lead compounds of anti-inflammatory and antitumor.This dissertation reviews the preliminary researches on the chemical constituents and pharmacological activities of Ajuga plants,thus establishing the basis for the comprehensive development and utilization of Ajuga.The chemical constituents were separated and purified by silica gel column chromatography,Sephadex LH-20 gel column chromatography,ODS column chromatography and semi-preparative high performance liquid chromatography.The structure was identified by spectroscopic methods such as HRMS、NMR、IR、ECD、X-RAY et al.Our results showed that 30 kinds of compounds including two new abietane-type diterpenoids 1~* and 2~* were isolated from the ethyl acetate part of Ajuga ovalifolia.The types of the compounds include flavonoids,steroids,diterpenes,iridoids,glycosides,phenolic acids,etc.Compounds 1~*-4,10,27,28 are abietane-type diterpenoids,and 11 is clerodane-type diterpenes.Compounds 6,14,16,22,29 are steroids.Compounds 8,13,15 are flavonoids.Compounds 26 is iridoid.Compounds 9,12 are coumarins.Compounds 5,7,18,20 are phenols.Compounds 17,19,21,23,24,25,30 are glycosides.Compounds 1~*-3,5,7,9,12,17-24,26,and 30 were isolated firstly from the Ajuga genus,and compound 4、8、10-11、14、16、25、27-29 were isolated from this plant for the first time.In addition,anti-inflammatory and anti-tumor pharmacological activities of some diterpenoid compounds were detected in many inflammatory and cancer cell lines.Our results showed that compound 4(ajuforrestin B)has good anti-tumor pharmacological activity and compound 10(ajudecumins A)has good anti-inflammatory and anti-tumor pharmacological activity.At last,the dissertation reviews the research progress on the chemical constituents and pharmacological activities of the Ajuga genus.Based on the collation and summary of the literature from 1922 to 2019,the chemical composition and pharmacological activity data of the Ajuga genus were improved,which provides the reference for the in-depth study of the Ajuga genus. |
