Background: PARP10 is a mono-ADP-ribosyltransferase.As an important regulatory factor in DNA and cancer biology,PARP10 can participate in DNA damage repair,promote cell proliferation,and play an important role in immune,metabolic,cell signal transduction,tumorigenesis and other biological processes.Overexpression of PARP10 has been found in most tumors,suggesting that it may promote cell transformation as an oncogene.Levels of PARP10 expression have been found to vary widely among tumor cell lines,but there is no conclusive evidence of PARP10’s role in specific tumors.Up to now,domestic and foreign reports on the relationship between PARP10 and oral squamous cell carcinoma(OSCC)are rare,and the specific role of PARP10 in the development of oral squamous cell carcinoma is unclear.Objective: To clarify the expression of PARP10 in oral squamous cell carcinoma,and to explore the correlation between PARP10 and the clinicopathological characteristics of oral squamous cell carcinoma,as well as the influence of PARP10 on the proliferation,invasion and migration of oral squamous cell carcinoma.It provides a theoretical basis for the diagnosis and evaluation of oral squamous cell carcinoma,and provides a new and more comprehensive direction for its treatment.Content: 1.The expression level of PARP10 in oral squamous cell carcinoma was detected,and its correlation with clinical pathological features was analyzed.2.By down-regulating PARP10,the effects of PARP10 on the proliferation,invasion and migration of oral squamous cell carcinoma cells were detected,and the mechanism of the abnormal expression of PARP10 on the occurrence and development of oral squamous cell carcinoma was studied.Methods : 1.Collect clinically diagnosed cases of oral squamous carcinoma in 92 cases,normal oral epithelial tissue in 11 cases and 34 cases of lymph node metastasis of oral squamous carcinoma tissue,at the organization level using immunohistochemical staining method to detect PARP10 expression in oral squamous cell carcinomas,using SPSS statistical analysis PARP10 expression and clinical pathological features.2.At the protein level,Western blot was used to detect the expression of PARP10 in various oral squamous cell carcinoma cell lines,and the PARP10 highly expressed oral squamous cell lines were screened out.The expression of PARP10 was down-regulated by si RNA for subsequent experiments.4.Western blot,cell proliferation test,immunofluorescence test,Transwell test and scratch test were used to detect the effect of PARP10down-regulation on the expression level,proliferation ability,morphology,invasion and migration ability of the related proteins of oral squamous cell carcinoma cells.Results: 1.PARP10 is highly expressed in oral squamous cell carcinoma tissues,but low in normal oral squamous epithelial tissues(P<0.01).2.The level of PARP10 expression is closely related to the histological differentiation,lymph node metastasis,pathological grading and other clinical and pathological features of oral squamous cell carcinoma patients,and positively correlated with the invasion and metastasis related proteins VEGF,MMP2,and cell proliferation active marker ki-67(P<0.05).3.HSC3 cells with high PARP10 expression were screened from four oral squamous cell carcinoma cells for sirna-parp10 transfection.After transfection,cell proliferation and division were reduced,apoptotic cells were significantly increased,cell morphology was changed,and polykaryotic macrokaryotic cells were increased.4.The scratch test showed that the wound healing rate of the transfection group was significantly lower than that of the control group(P<0.01).5.The Transwell experiment found that the number of cells migrating or invading to the small outdoor side in the transfection group was significantly less than that in the control group(P<0.01).The expression of invasion and migration related genes,VEGF,MMP2,N-cadherin,Snail,Slug,Vimentin,Wnt5a/b,and beta-catinen decreased,while the expression of E-cadherin increased.The expression of related pathway proteins p-Akt,p-P38,p-RSK,p-SRC decreased.6.Cell proliferation experiments showed that cell proliferation was significantly reduced in the transfection group,the expressions of pro-apoptotic genes Bax,cleaved Caspase-3,Caspase-3 were increased,and the expressions of anti-apoptotic genes Bcl-XL and Bcl-2 were decreased.The expression of Cyclin B and Cyclin D decreased,and the expression of tumor suppressor genes p53 and Cyclin inhibitors p21 and p27 increased.Conclusion: 1.PARP10 is highly expressed in oral squamous cell carcinoma and is closely related to histological differentiation and pathological grading.2.The expression level of PARP10 is closely related to the proliferation,invasion and migration of oral squamous cell carcinoma,and is involved in the occurrence and development of oral squamous cell carcinoma.Conclusions: 1.PARP10 is highly expressed in oral squamous cell carcinoma and is closely related to histological differentiation and pathological grading.2.PARP10 is involved in the occurrence and development of oral squamous cell carcinoma,and its expression level is closely related to the proliferation,invasion and migration of oral squamous cell carcinoma. |
