Objective: To investigate the effects of ART3(mono-ADP-ribosyltransferase3)gene on proliferation, apoptosis,invasionof breast cancer cells (MDA-MB-231).Methods:Human breast cancer cells (MDA-MB-231) as a model andtransfected with the eukaryotic cell recombinant plasmid pCMV-ART3–myc-tag.Using G418to screen stable transfectants and Western blot toverification the expression of ART3.The proliferation tested byMTT,apoptosis tested by flow cytometry analysis with Annexin-V and PIdouble staining,and invasion in Transwell assay.Results:The ART3gene stably overexpressed in breast cancer cells(MDA-MB-231).ART3overexpression group cells with higher proliferationand invasiveness had lower apoptosis rate compared to non-transfectedbreast cancer cells.Conclusions: The experiment successfully built ART3overexpressionstable clones, and found ART3can promote the proliferation, invasion andinhibit apoptosis of breast cancer cells.Also the experiment had laid for further study of the role of ART3in the tumorigenesis and development ofbreast cancer. Objective: To investigate the effects of ART3(mono-ADP-ribosyltransferase3)gene on tumor growth in nude micexenograft models of MDA-MB-231cells.Methods: Using the ART3overexpression and non-transfectedMDA-MB-231breast cancer cells to build nude mice xenograftmodels.After tumor formation, the tumor volume measurement were doneevery3days,and the tumor growth curve was drawn.The mice weresacrificed after30days,and the tumor tissue were extracted for western blotto detect ART3exprssion.Results: The nude mice xenograft models were successfully constructed,and the tumors of ART3overexpression group grew faster thanthat of non-transfected group.The tumor tissue protein western blotdisplayed higher ART3expression in ART3overexpression xenograft thanthat in non-transfected xenograft.Conclusions: The experiment successfully built nude mice models ofhuman breast cancer cells, and found ART3can promote tumor growth ofxenograft models,also the experiment had laid for further study of the roleof ART3in vivo on tumorigenesis and development of breast cancer.
