Abstract/Summary:
Objective: To explore the ectopic osteogenesis effect and mechanism of the porous hydroxyapatite(HA)ceramic scaffolds as the artificial bone substitute materials.Methods: Co Cultured human placenta derived mesenchymal stem cells with porous HA ceramic scaffolds for 7 days,cells adherent to the porous HA ceramic scaffolds are taken out in the Sterile environment to implanted in the beagle’s back muscles for the experimental group,And implanted blanket porous HA ceramic scaffolds in the beagle’s back muscles for the control group,respectively remove the ectopic osteogenesis organization at 4,8,12 weeks after implantation,then detect and analysis the osteogenesis effec,gene transcriptome sequencing,RT-PCR and Western Blot of the ectopic ossification organizations.Results: Tissue section dyeing results showed that the implanted at 12 weeks of Heterotopic osteogenesis has a large number of new blood vessels and new bone tissue form,and experimental group is obviously beteer than the control group.The sequencing results showed that the heterotopic ossification process involving multiple signaling pathways,mainly for the TGF-β and BMP signaling pathways.The results of RT-PCR and Western Blot were consistent with the transcriptome sequencing results.Conclusion: Porous HA scaffolds have good ectopic bone formation in vivo,and the human placenta derived mesenchymal stem cells can promote the formation of new bone,The osteorelated pathways that may be involved in the ectopic osteogenesis are TGF-beta and BMP signaling pathways as well as the osteogenic related genes involved in BMPER,RUNX2,COL-1,VEGFA,BMP2/4/6,TGFB.
