Objective of the experiment:To find a natural and simple and effective way to improve sleep disorders,we used inhalation,oral and Shenque(navel)administration to study the effects of lavender oil on sleep disorders.Experimental method:Determination of the upper and lower doses of pentobarbital sodium in test animals(mouse,rat);the two animals(mouse,rat)were divided into lavender oil oral group and inhalation group 10 rats in each group were treated with umbilical administration group,positive control group and blank group.The sleeping rate,sleep latency,and sleep duration were measured in each animal group tested.Bioavailability study of oral administration of lavender essential oil and Shenque points:SD Twelve rats were randomly divided into groups.Each group consisted of 6 oral groups(Linalool)and Shenalool Shenque.The doses were 800 mg/kg.The time points after administration(5,10,15,20)Blood was collected from the venous region at the time of 30,40,50,60,120,180,240,360 and 480 min.The concentration of linalool,the main component of lavender in plasma,was determined by high performance liquid chromatography and calculated using DAS 2.0 software.Pharmacokinetic parameters to evaluate the relative bioavailability of Linalool Shenque for Linalool.Experimental results:subthreshold dose 30 mg/kg,supraminal dose 60 mg/kg in mice,subthreshold dose 20 mg/kg in rats,suprathreshold dose 40 mg/kg;sleep rate test:two animals(mice,There was a significant difference between the navel-administered group and the blank group(P<0.05).The sleeping rate of the navel-administered group was the same as that of the positive drug in the navel-administered group,and was superior to that of the inhalation group and the oral group,respectively;sleep latency Experiment:There was a significant difference between the navel-administered group and the blank group in both animals(mice and rats)(P<0.05).The incubation period was the shortest in the navel-administered group and the positive-administered group,and the navel of the rats was the shortest.The sleep latency of the drug-administered group was slightly shorter than that of the positive drug group,and the next two animals were successively better than the inhalation group and the oral group;Sleep duration:the difference was significant between the navel-administered group and the blank group(P<0.05).Animals in the navel administration group were superior to the positive drug group,inhalation group and oral group successively;relative bioavailability:The linear range of the mass concentration of linalool,the main component of lavender,was(45.45-1818.18 ng·m L-1)..The p1/2 pharmacokinetic parameters of rats in Linalool and Linalool Shenque were 107.59 and 107.59,MRT(min)was 46.86 and 149.64,and Cmax(ng·m L-1)was 2065.24 and 2382.45,respectively.The relative bioavailability of Linalool Shenque was 343.14%.The relative bioavailability of Linalool Shenque was calculated as AUC0-t,and the AUC0-t(ng·min·mL-1)were 104469.31 and 358472.97,respectively.The conclusion of the experiment:The administration of lavender essential oil Shenque acupoint is a simple,fast and effective way to improve sleep quality and improve sleep disorders.The umbilical effect of improving sleep is greater than that of inhalation and oral administration;and the effect of improving sleep disorders is comparable to that of the positive drug Diazepam.Umbilical administration has higher bioavailability than oral administration.
