The fruits we studied were from Buddleja lindleyana Fort.,Loganiaceae.Triterpenes,flavonoids,phenethanol glycosides,iridoids,and other constituents were confirmed in The fruit of Buddleja lindleyana Fort.according to our previous studies.Among them,triterpenes were revealed with positive neuroprotective effect.In this paper,the constituents of the water fraction were separated and purified in order to enrich the chemical constituents in the fruit of B.lindleyana Fort.,which neuroprotective effect was screened furthermore.Objective:To enrich the chemical constituents in the fruit of B.lindleyana Fort.with the study of the water fraction.And the neuroprotective effect of some constituents was screened in order to find the valuable ones with positive effect.Methods:Multiple column chromatographies were applicated in separating and purifying,and the structure of the certain constituents were identified with modern spectroscopic technology.Then SH-SY5Y cellular models were adopted to screen positive neuroprotective effect between some of the constituents.Results:19 compounds were isolated and purified from B.lindleyana Fort.,and13 of them were identified as mimengoside C(1),isoacteoside(2),apigenin-7-O-β-D-rutinoside(3),acteoside(4),4′-hydroxyphenyl ethyl vanillate(5),acacetin-7-O-β-D-glucoside(6),6-O-vanilloyl-ajugol(7),syringaresinol-4’-O-β-D-glucopyranoside(8),linarin(9),eleutheroside B(10),coniferin(11),buddlejasaponinⅣ_b(12),6-O-(3"-O-p-coumaroyl-α-L-rhamnopyranosy)catalpol(13).Compounds 1,7,8,and 10 were tested for their positive neuroprotective effect on SH-SY5Y cellular models.The results of MTT assay showed that all the above compounds could significantly increase the cell survival rate.Conclusions:Among the compounds above,compounds 2,4,5,8,10,11,and13 were firstly isolated from the fruit of B.lindleyana Fort.,while compounds 5,8,10,11,and 13 were firstly isolated from this genus.The results of MTT assay showed that compounds 1,7,8,and 10 possess positive neuroprotective effect on SH-SY5Y cellular models.
