Objective:To inverstigate the effects of eukaryotic translation elongation factor 1A1(eEF1A1)on chemotherapy sensitivity of hepatocellular carcinoma cells by mediating ISGylation.Methods:We detected the mRNA and protein expression of eEF1A1 by RT-PCR and Western blot technique and ensured the lever of ISGylation in hepatocellular carcinoma cell lines.A hepatocellular carcinoma cell line with stable low expression of eEF1A1 was built through short hairpin RNA interference technology,and the lever of ISGylation,cell growth ability and cell apoptosis were detected after Adriamycin treatment.The hepatocellular carcinoma cell line with stable low expression of eEF1A1 were restored eEF1A1 expression,and then the lever of ISGylation,cell growth ability and cell apoptosis were detected after Adriamycin treatment.Results:It was found that the expression of eEF1A1 was high and the level of ISGylation was low among hepatocellular carcinoma cell lines.After treatment of Adriamycin,compare to negative control group,the lever of ISGylation increased and the cell growth ability decreased significantly(P<0.01)and the proportion of apoptotic cells increased evidently(P<0.05)in the MCC97H with stable low expression of eEF1A1.After treatment of Adriamycin,compare to negative control group,the lever of ISGylation decreased and the cell growth ability increased significantly(P<0.01)and the proportion of apoptotic cells decreased evidently(P<0.05)in the MCC97H with over expression of eEF1A1.After treatment of Adriamycin,compare to MCC97H with stable low expression of eEF1A1,the lever of ISGylation decreased and the cell growth ability increased significantly(P<0.01)and the proportion of apoptotic cells decreased evidently(P<0.05)in the steady transfacted MCC97H whose expression of eEF1A1 was recovered.Conclusion:In hepatocellular carcinoma cells,eEF1A1 can affect the biological characteristics of HCC cells in Adriamycin treated environment by regulating the level of ISG.
