| Objective:To explore the mechanism of eukaryotic translation elongation factor 1a1(eEF1A1)regulating the expression of NIN1/PSMD8 binding protein homologue(NOB1)on invasion and metastasis of hepatocellular carcinoma.Methods:1.Detect the mRNA and protein expressions of eEF1A1 and NOB1 in various Hepatocellular Carcinoma(HCC)cell lines(MHCC97h,HCCLM3,SMMC7721 and Hep3B)and normal liver cell HL7702 were detected,and the correlation between the expressions of the two was analyzed.2.To observe the changes of invasion and migration ability of HCC cells after interfering with and up regulating the expression of eEF1A1 by Transwell invasion and migration experiment and RTCA experiment.3.The changes of NOB1 mRNA and protein expression were observed by real-time fluorescence quantitative PCR and Western blot after changing the expression of eEF1A1 in HCC cells.4.The expression of NOB1 was over expressed in HCCLM3 of HCC cells,which stably interfered with the expression of eEF1A1,or in MHCC97 H of HCC cells,which over expressed eEF1A1.The expression level of eEF1A1 and NOB1 and the ability of invasion and migration of cells were observed.Results:1.The mRNA and protein expressions of eEF1A1 and NOB1 in HCC cells are significantly higher than those in normal liver cells.2.After interfering and up-regulating the expression of eEF1A1 in liver cancer cells,we found that the invasion and migration ability of HCC cells were weakened and enhanced respectively.3.After changing the expression of eEF1A1 in HCC cells,it was found that eEF1A1 and NOB1 were positively correlated in mRNA and protein expression levels.4.eEF1A1 depends on NOB1 to regulate invasion and metastasis of HCC cells;Conclusion:In hepatocellular carcinoma,eEF1A1 positively regulates the expression of NOB1,thus affecting the invasion and migration ability of HCC cells. |
