Pandanus tectorius Soland.is a common semi-mangrove at estuary area,it belongs to genus Pandanus,and it grown on the edge of the intertidal zone mainly.In china,it is mostly founld in Hainan,Fujian,Guangxi.It’s a traditional medicine in south of the Five Ridges area of China,currently,it is received in the "local Chinese herbal medicine standards of Guangdong Province "(2011 Edition),with the function of calming the liver and clearing heat,relieving pain,reducing blood fat,it is usually used to treat hepatitis and hyperlipidemia.In the prophase study,when we study the lipid-lowering activity of the fruit of P.tectorius Soland.,we find that its stem bark has good lipid-lowering activity either.However,there is still no reports about it’s lipid-lowering active.So we analysis the chemical constituents of the stem bark,and screened the lipid lowering activity compounds,to know the material basis of its lipid lowering activity,preparing for the utilization of P.tectorius Soland.comprehensively.In the thesis,with modern separation technology and chromatographic methods,the extraction of ethyl acetate and n-butanol from the bark of P.tectorius Soland.are systematically studied,twenty-four compounds from the bark of P.tectorius Soland.have been identified in our studies.Isolation and identification of 24 compounds from the stem bark of P.tectorius Soland.in the 75%ethanol extraction,their structures were identified as pinoresinol(1),3-hydroxy-2-isopropenyl-dihydrobenzofuran-5-carboxylic acid methyl ester(2),Benzyl alcohol-β-D-glucopyranoside(3),epi-pinoresinol(4),Salicifoliol(5),Loliolide(6),Pinoresinol-4-O-β-D-glucopyranoside(7),erythro-2,3-bis(4-hydroxy-3-methoxyphenyl)-3ethoxypropan-l-ol(8),thero-2,3-bis(4-hydroxy-3-methoxyphenyl)-3-ethoxypropan-l-ol(9),Eudesmin A(10),Protocatechuic acid(11),caffeic acid(12),trans-p-hydroxycinnamic acid(13),Ferulic acid(14),1’-O-benzyl-α-L-rhamnopyranosyl-(1"-6’)-β-D-glucopyranoside(15),dihydrodehydrodiconiferyl alcohol(16),Isovitexin(17),Vitexin(18),3,5-dicaffeoylquinic acid methyl ester(19),3,5-dicaffeoylquinic acid ethyl ester(20),3,4-dicaffeoylquinic acid ethyl ester(21),(+)-lyoniresinol 3a-O-β-glucopyranoside(22),(-)-lyoniresinol 3a-O-βglucopyranoside(23),chlorogenic acid(24).The above compounds were first time isolated to the bark of P.tectorius Soland..Additionally,In our work,we tested the activity of lipid-lowering of the compounds which were from the bark of P.tectorius Soland.,and their were.The extraction of compounds 7 and 24 from P.tectorius Soland.were screened for the activity of lipid-lowering in HepG2 cells elicited by oleic acid.The effect of Pinoresinol-4-O-β-D-glucopyranoside and chlorogenic acid on inhibiting lipid accumulation which were stimulated by Oil-acid in HepG2 cells were studied by microscope and spectrophotometry.The intracellular levels of total cholesterol(TC)and triglyceride(TG)were quantified by specific kits.The results showed that:(1)The compounds 7 and 24 of the bark from Pandanus tectorius Soland.can significantly inhibit the accumulation of lipid,but the other compounds which were isolated from P.tectorius Soland doesn’t have the activity;(2)The compounds 7 and 24 not only can be decreased lipid accumulation,but also reduce the intracellular TC and TG at the concentration of 50 μmol/L.Above all,systematical analysis of the chemical constituents from the bark of Pandanus tectorius Soland.has been conducted in the thesis.Furthermore,the activity of lipid-lowering of chemical composition isolated from the bark of P.tectorius Soland.were studied.The above research can be the basis for further more study of Pandanus tectorius Soland..
