Methods(1)Western blot was used to investigate the LAMP3 expression level in 16 cases of human bladder cancer tissues and paired ANT.(2)Real-time PCR was used to investigate the.expression of LAMP3 mRNA in 32 cases of human bladder cancer tissues and paired ANT.(3)Immunohistochemistry was utilized to detect the expression of LAMP3 and Ki67 protein in 132 cases of bladder cancer speci mens.(4)Kaplan-Meier survival curve was used to investigate the association of LAMP3 protein expression with the prognosis of the patients with bladder cancer.(5)Statistical analysis:All statistical analyses were conducted using SPSS version 20.0 statistical software,all data were expressed as x±s.Comparisons between groups for statistical significance were performed with a 2-tailed paired Student’s t test.The relationship between LAMP3 expression and clinicopathologic parameters was analyzed by the χ2 test.Survival curves were plotted using·the Kaplan-Meier method and survival data were evaluated by univariate and multivariate cox regression analyses.A P value less than 0.05 was considered as statistical significance.Results(1)Western Blot results showed that 15 of 16 patients had a higher LAMP3 protein expression level compared with paired ANT samples.(2)Real-time PCR showed that 30 of 32 patients had a higher LAMP3 mRNA expression level compared with paired ANT samples,and the differences were statistical significances.(3)Immunohistochemistry results showed that 70 cases had LAMP3 low expression and 54 cases with high expression of the 134 patients with bladder cancer.Statistical analysis revealed that the LAMP3 levels were associated with the grade,T classification or multiplicity classification,but they were not associated with age.(4)Kaplan-Meier survival curves showed that the overall survival,cancer-specific survival and progression-free survival of patients with low levels of LAMP3 were significantly better than that for patients with high levels of LAMP3,and the differences were statistical significances(P<0.05).Univariate and multivariate analyses indicated that gender,N classification,and the expression of LAMP3 were prognostic factors.
