| Objective To investigate the effect of radix tetrastigme,taxus and their combination on myeloid-derived suppression cells(MDSCs)and related cytokines of lewis tumor bearing C57BL/6 mice,explore and compare the probably mechanism of reversing immunosuppression,and provide the experimental evidence for clinical use.Methods ①The mice model of Lewis tumor were establish by subcutaneous inoculation and randomly divided into 4 groups:radix tetrastigme group,taxus group,combination group and normal saline(NS)group.② Irrigation once per day for model groups 24 hours after inoculation and free diet for the normal group.Observe the change of mental state,weight,tumor and mortality of each group every day.Calculate the inhibitory rate of tumor after 14 days of irrigation.③Collect eyeball blood after 14 days of irrigation.Execute the mice by removing cervical and take out their spleen and tumor.ELISA for measuring COX-2,VEGF,Arg-1 and iNOS in peripheral blood,flow cytometry for the proportion of MDSCs in spleen and immunohistochemistry for Arg-1,NOS expressed by tumor tissue.④SPSS 17.0 is used for statistical analysis by ANOVA.P<0.05 was considered statistically for differences.Results ①Weight:It has significant difference between group NS and TCM and has no between group TCM.②The inhibitory rate of tumor:In the order from high to low is group radix tetrastigme(53.99%),group combination(41.31%),group taxus(23.72%)by the volume of tumor and group radix tetrastigme(52.32%),group combination(31.29%).group taxus(24.34%)by the weight of tumor.③Content of COX-2 in the peripheral blood:It has notable significant difference between group NS and TCM(P<0.01).Group radix tetrastigme and combination has significant difference between taxus(P<0.05).but they have nothing difference between each other(P>0.05).④Content of VEGF in the peripheral blood:Group NS has notable significant difference between group radix tetrastigme and conbination(P<0.01)and no difference between taxus(P>0.05).It has no significant difference between Group radix tetrastigme and combination(P>0.05).⑤Content and expression of Arg-1:In the peripheral blood and tumor,group NS has significant difference between group radix tetrastigme(P<0.01)and combination(P<0.05),and no difference between taxus(P>0.05),also has no significant difference between Group radix tetrastigme and combination(P>0.05).It has significant difference between group radix tetrastigme and combination(P<0.05)in peripheral blood while not in the tumor(P>0.05).⑥Content and expression of i-NOS:In peripheral blood,group NS has significant difference between group radix tetrastigme and combination(P<0.01),and no difference between taxus(P>0.05),while it has between radix tetrastigme and combination(P<0.01).In tumor,group NS has significant difference between TCM(P<0.05).Group radix tetrastigme and combination has similar affection(P>0.05),but much difference between taxus(P<.05).⑦The proportion of MDSCs in spleen:It has notable significant difference between group NS and TCM(P<0.01).Group radix tetrastigme and combination has nothing different(P>0.05),but they all have significant difference between taxus(P<0.05)Conclusion ① The traditional Chinese medicine,radix tetrastigme,taxus and their Combination have anti-tumor effect.② Radix tetrastigme and their combination can downregulate the content of COX-2、VEGF in peripheral blood notablely while taxus has significant effect only in COX-2.③Radix tetrastigme and their combination can inhibit the high-expression to Arg-1,iNOS of MDSCs while taxus has no significant effect.④Radix tetrastigme,taxus and their combination can reduce the proportion of MDSCs in spleen of mice bearing Lewis tumor significantly,helping to improve the immune suppression. |
