Myeloid Derived Suppressor Cells In Peripheral Blood Of Multiple Myeloma

Abstract/Summary:
Objective: Myeloid derived suppressor cells(MDSCs)are a group of myeloid derived heterogeneous cell populations with immunosuppressive function.In recent years,some studies found that MDSCs were obviously up-regulated in patients with a variety of solid tumors in peripheral blood.Moreover,MDSCs have a potential immunosuppressive effect in a variety of solid tumors.But its expressions and functions in myeloma are still not clear.In this study,we investigated the proportion of MDSCs and other immune cells in peripheral blood of patients with myeloma and the m RNA expression of MDSCs related immunosuppressive factors.Methods: 1.The clinical data of the patients were analyzed,and a total of 32 patients with newly diagnosed myeloma were selected as the experimental group,and a total of 20 healthy persons were selected as normal group.To determine the type of immunity,the experimental group was analyzed by immune fixation electrophoresis.Then,the concentrations of globulin and C-reactive protein in the plasma were detected from the experimental group and the control group respectively.The total numbers of white blood cells in peripheral blood from experimental group and control group were observed by blood cell analyzer.2.The proportions of T lymphocyte subsets(CD4+CXCR5+ICOS+Tfh cells,CD4+CD25+CD127low/-Treg cells and CD8+Tc cells,CD4+Th cells),CD19+B lymphocytes and CD16+CD56+NK cells in experimental and control groups were analyzed using flow cytometry.3.The percentages of CD11b+CD33+CD14-MDSCs,CD11b+CD33+CD14-CD15+PMN-MDSCs,CD11b+CD33+CD14-CD15-M-MDSCs in the peripheral blood from the experimental group and the control group were detected with flow cytometry.In two groups,the m RNA expressions of ARG1,i NOS and VEGF which related with the functions of MDSCs were quantified by QRT-PCR.Results: 1.Multiple myeloma(IgG)is the most common type of bone marrow tumor.The concentrations of immunoglobulin and C reactive protein in the experimental group were obviously higher than the control group,and the total numbers of peripheral white blood cell were no significant difference between the experimental group and the control group.2.Compared with the normal group,the proportions of CD4+CD25+CD127LOW/-Treg and CD4+CXCR5+ICOS+Tfh cells were increased significantly in the experimental group.In the experimental group,the proportion of CD4+Th was slightly reduced,and the proportion of CD8+Tc was apparently increased,resulting in ratio of Th and Tc cells was significantly lower than the control group.The percentage of CD19+B lymph was slightly up-regulated in the experimental group,and the proportion of CD16+CD56+NK cells was lower than the control group,but there is significant difference.3.The proportions of CD11b+CD33+CD14-MDSCs,CD11b+CD33+CD14-CD15+PMN-MDSCs in the peripheral blood of the experimental group were significantly higher than the normal group,while the proportions of CD11b+CD33+CD14-CD15-M-MDSCs in the peripheral blood were no significantly difference between the experimental group and the control group.The relative m RNA expression levels of ARG1,i NOS and VEGF in the peripheral blood of the experimental group were significantly higher than those of the control group.Conclusions: More PMN-MDSCs and higher levels of MDSCs relative factors,which contributed to inhibit in the proliferation and activation of T cells were detected in the peripheral blood of patients with MM.
