Objective By exploring the effect of Zhongyaoqubanshuang on SOD,MDA a nd melanin granules in the guinea pigs model of pigmentation after UVB irrad iated,and researching Zhongyaoqubanshuang’s possible mechanisms on the tre atment of skin pigmentation,to build the experimental basis for clinical applic ation and providing new ideas and methods for traditional Chinese medicine ex ternal treatment on skin pigmentation.Methods 50 qualified male guinea pigs with brown skin were randomly divided into five groups:normal control group,model group,Qingkun group,high-density Zhongyaoqubanshuang group and low-density Zhongyaoqubanshuang group.After all guinea pigs epilation on their back,except the normal control group,the remaining four groups were irradiated with UVB on their brown-skin back,irradiated every 60min,qd.After four weeks of continuous irradiation,except the normal control group,model group,Qingkun group,high-density Zhongyaoqubanshuang group and low-density Zhongyaoqubanshuang group were given a single base cream,Qingkun cream,highdensity Zhongyaoqubanshuang and low-density Zhongyaoqubanshuang on skin lesions,every 5min to massage qd,continuously for 4 weeks.All guinea pigs were killed after four weeks’ treatment.The normal control group were taken the sklin on their back,the other four groups were taken skin lesions who were radiated under UV.All the skin specimens were fixed in the 10%formalin.Then examine SOD and MDA,and observe melanin granules by microscope through HE staining and ferrous sulfate staining.Measure the average IOD of melanin granules by immunohistochemistry.Then analyze the positive expression level of melanin granules.Results 1.Eye-observation:except the normal control group,the other guinea pigs groups appears successful skin pigmentation after UVB irradiation The ski n pigmentation of guinea pigs among Qingkun group,high-density Zhongyaoqu banshuang group and low-density Zhongyaoqubanshuang group reduced after tre atment.2.SOD activity in each treatment group and model group were signifi cantly lower than normal control group,MDA value was significantly higher th an the normal control group(P<0.05).SOD activity in Qingkun group,high-de nsity Zhongyaoqubanshuang group and low-density Zhongyaoqubanshuang group were significantly higher than model group,MDA value was significantly low er than model group(P<0.05).SOD activity in low-density Zhongyaoqubanshua ng group were significantly higher than Qingkun group and high-density Zhong yaoqubanshuang group,MDA value was significantly lower than Qingkun grou p and high-density Zhongyaoqubanshuang group(P<0.05).There was no statisti cal difference between the Qingkun group and high-density Zhongyaoqubanshua ng group(P>0.05).3.Results of HE staining,ferrous sulfate staining and imm unohistochemistry:compared with the normal control group,melanin granules in other groups located in the basal layer of skin were significantly increased,the expression of melanin granules were significantly higher than the normal c ontrol group(P<0.05).Melanin granules in Qingkun group and high-density Zho ngyaoqubanshuang group,and low-density Zhongyaoqubanshuang group were si gnificantly decreased than model group,and the expression of melanin granules were significantly decreased(P<0.05).Melanin granules in low-density Zhongy aoqubanshuang group were significantly lower than Qingkun group and high-de nsity Zhongyaoqubanshuang group,and the expression of melanin granules wer e significantly lower(P<0.05).There was no statistical difference between the melanin granules in the Qingkun group and high-density Zhongyaoqubanshuang group,and the expression of melanin granules were also no difference(P>0.05).Conclusion(1)Building animal model of pigmentation which use UVB to irradiat on pigmentation male guinea pigs with brown hair is successful.(2)Zhongyaoqubanshuang has a significant improvement on pigmented skin lesions of guinea pigs.The treatment mechanism maybe associated with the enhancement of SOD activity,the reduction of MDA value and the inhibition of melanin granules generated(3)The effect of Zhongyaoqubanshuang in low-density was better than Qingkun cream and high-density Zhongyaoqubanshuang(4)On the short-term,the high-density Zhongyaoqubanshuang has the same effect as Qingkun cream,but Qingkun cream has a stronger stimulation.
