Objective To investigate the expression and correlation of micro RNA-26 a,143,195 in tissues and matching sera of patients with sporadic/familial breast cancer(BC)and to find new promising potential biomarkers for BC.Methods Tumor tissues,adjacent non-tumor tissues and matching sera were collected from patients with familial BC and patients with sporadic BC.Normal control serum samples were collected from healthy subjects.Total RNAs were extracted from all these samples and the aberrant expression levels of a panel of 3mi RNAs(mi RNA-26 a,143,195)were quantified by using real-time PCR technique.Wilcoxon test,Mann-Whitney test and t-test were used for statistical analysis.Results mi RNA-26 a,143,195 were significantly down-regulated in the tumor and serum specimens compared with those in normal controls(P < 0.05).mi RNA-26 a,143were under-expressed in familial BC tissues compared with sporadic BC tissues(P<0.05),whereas mi RNA-143,195 were under-expressed in sera of familial BC compared with sporadic BC(P<0.05).Furthermore,a high correlation of mi RNA-143 expression level was found between BC tumors and sera(r=0.545,P<0.05).Conclusions This study highlights the roles of mi RNAs in BC diagnosis.mi RNA-143 could be promising potential blood-based biomarker for familial BC diagnosis.
