The mechanism of breast cancer is of the barriers to the mechanism of apoptosis caused by abnormal cell proliferation.The incidence of breast cancer is a multi-stage,mutifactor,multiple genes are involved in the process,the specfic cause and mechanism is not yet very clear.Expression of Skp2(S phase kinase associated protease2) newly discovered Skp-Cullin,the F-box (SCF),the E3enzyme complexes in an F-box protein,specifically recognized in the ubiquitin-proteasome pathway substratesinvolved in the regulation of cell proliferation and appoptosis,and closely related to tumor development.p27Kipl (cell cycle inhibitory protein of p27Kip1) is the cyclin-dependent kinase inhibitor discovered in recent years, which is named expression of cyclin-dependent kinase inhibitors,and which is one of the numbers of the family of the CDI,and it can prevent the cells through the G1/S inhibition of cell proliferation.These two important signaling molecule involved in the regulation of cell proliferation and apoptosis,closely related to tumor development,has become one of the past few years the research focus of tumor mechanism.But it is rarely reported in the development of breast cacer research.ObjectiveDetection of Breast Cancer Organization of Skp2and p27Kipl mRNA and itsprotein expression.to explore in breast cancer development mechanism,and also discussed the relationship betweem the two clinical cases factors.MethodsSemi-quantitative reverse transcription polymerase chain recation(RT-PCR) was used to detect the mRNA differential expression of Skp2and p27kip1among40cases of breast cancer,20cases of breast fibroadenoma tissue and20cases of normal breast tissue Furthermore, the relationship between mRNA expression and clinical pathological factors was analysised. Immunohistochemical(PV-6000) two-step detection of Skp2and p27kipl protein expression in40cases of breast cancer and to study their expression levels and clinicopathologic factors in20cases of normal breast tissue as controls.RusultsThe average mRNA expression of Skp2was significantly higher in breast cancer than that in breast fibroadenoma and normal tissue, but that in normal breast tisssue and fibroadenoma was no significant difference. The average mRNA expression of p27kipl was significantly lower in breast cancer than that in breast fibroadenoma and normal breast tissue, but that in normal breast tissue and fibroadenoma was no significant difference. The mRNA expression of Skp2was positively related to pathological grade, lymphatic metastasis, but were irrelevant to patients’ age,tumor size and surgical pathological stage. The mRNA expression of p27kip1was negatively related to pathological grade, and lymphatic metastasis, but was irrelevant to patients’age, tumor size and asurgical pathological stage. The mRNA expression of Skp2was negatively related to that of p27kipl mRNA in breast tissue.Skp2protein expression in breast cancer tissue levels and histological grade,lymph node metastasis were positively correlated;of p27kipl protein expression and histological grade,lymph node metastasis was a negative correlation. But both of which was irrelevant to patients’age,tumor size and surgical pathological stage. Skp2and p27kipl proteins in breast cancer tissue was negatively correlated. ConclusionsThe high expression of Skp2and low expression of p27kip1in breast cancer tissue involved in breast carcinogenesis,development process.The two indicators in the joint detection of gene and protein levels may help predict breast prognosis and treatment of breast cancer has some guding significance.
