| Curcuma wenyujin Y.H.Chen et C.Ling is a member of Curcuma genus,zingiberaceae family(ginger family),which is mainly distributed in the southern of Zhejiang province.It’s one of the "Zhe Bawei".This drug tastes spicy,bitter and cold.It has effect of unobstructing gas,circulation,eliminating irritability stagnation,conduciving gallbladder and removing jaundice.Its primarily for the treatment of amenorrhea,dysmenorrhea,abdominal pain,tingling,fever coma,epilepsy mad,red urine and jaundice and so on.So far,the research of the C.wenyujin has achieved many results.To the research of the chemical composition of C.wenyujin,there are more than one hundred compounds have been isolated and identified and some of them have good bioactivity.However,only Dong Jianyong has done some relevant reports on the chemical composition of C.wenyujin dreg after extracting volatile oil.In view of those research,in order to take full advantage of this medicine resource and further development of the this herb and turning waste into treasure,to separate the chemical composition of the C.wenyujin derg and researching the bioactivity of the isolated compounds has been a very meaningful work.In this study,Conventional separation methods like positive phase/reverse phase silica gel column chromatography,thin layer chromatography,Sephadex LH-20 gel column chromatography and modern separation techniques(HPLC)separation techniques were used to isolated and purified the chemical composition from the petroleum ether and ethyl acetate extracts of the ethanol extract of C.wenyujin dreg,and used physical and chemical properties and spectroscopic analysis and combined with the literature report.32 compounds(1-32)were isolated and identified from the C.wenyujin dreg,including 22 sesquiterpenes(1-22),8 diphenyl-heptanes(23-30)and two other compounds(31-32).4,8-Dioxo-6β-Hydroxyl7β,11-epoxy carabrane(1),4,8-Dioxo-6β-Hydroxyl-7α,11-epoxy carabrane(2),wenyujinins O-P(10-11)and wenyujinins Q-T(27-30)are new compounds.4,8-dioxo-6β-methoxy-7α,11-epoxycarabrane(4),1,5-epoxy-3-hydroxy-1-(4,5-dihydroxy-3-methoxyphenyl)-7-(4-hydroxy3-methoxyphenyl)hept-ane(23),1,5-epoxy-3-acetyl-1-(4,5-dihydroxy-3-methoxyphenyl)-7-(4hydroxy-3-methoxyph-enyl)heptane(24),rel-(3R,5S)-3,5-dihydroxy-1-(4-hydroxy-3-meth-oxyphenyl)-7-(3,4-dihydr-oxyphenyl)Hept-ane(26)and(S)-1-chloro-pentadecan-2-ol(32)were isolated from the Curcuma genus for the first time;4,8-dioxo-6β-methoxy-7β,11-epoxycarabrane(3),octahydro-5,8-dihydroxy-5,8α-dimethyl-3-(1-methylethylidene)(8),neoprocurcumenol(14),Phaeusmane G(21),rel-(3R,5S)-3,5-dihydroxy-1-(4-hydroxy-3-meth-oxyphenyl)-7-(4hydroxy-phenyl)heptane(25)were isolated from the C.wenyujin for the first time.The ethanol extract,petroleum ether extract and ethyl acetate extract were tested with antibacterial activity and Artemia lethal activity.The results showed that the petroleum ether extract and ethyl acetate extract showed activity against Escherichia coli;none of them showed Artemia lethal activity.Compounds 1-20 and 22-30 were tested with antibacterial activity.The results showed that 4,9,and 16 showed significant antibacterial activity against E.coli with the same MIC values of 1.25μg/mL,and 18 exhibited significant antibacterial activity against S.albus with the MIC value of 1.25μg/mL.In the meanwhile,we have extracted the residual oil of the Curcuma wenyujin dreg,and analysis the GS-MS composition and antibacterial activity of the residual oil.The results showed that the residual oil has strong antibacterial activity to the M.tetragenus,E.coli and S.aureus with the same MIC values of 44.4μg/mL. |
