| Curcuma wenyujin Y.H.Chen et C.Ling is a member of Curcuma genus, Zingiberaceae family (ginger family), which is distributed in Zhejiang Province. It have been found diverse pharmacological activities such as promising chemopreventive dietary agents exhibiting inhibition of cyclooxygenase and lipoxygenase, apoptosis induction, and anti-tumorigenic activities. In this thesis, chemical constituents from 95 % ethanol extract of C. wenyujin were studied. On the basis of different kinds of morden chromatography and identification methods, seventeen compounds were determined, most of them were sesquiterpenes and diterpenes. Their structures were identified as crotepoxide (1), 4-epi-curcumenol (2), curcumenol (3), curcumadiol (4), gweicurculactone (5), curcumin (6), demethoxycurcumin (7), bisdemethoxycurcumin (8), curcumrinol A-C (9-11), curcuminol D-E (12-13), curcuminol F-I (14-17) by physico-chemical propertities and spectral data, respectively. Compounds 9, 10,11, 12,13,14,15,16 and 17 were assigned as new compounds. Compound 1-5 were isolated from C. wenyujin for the first time and ambiguous or inaccurate 1H NMR and 13C NMR data of compound 5 in the literature were reassigned definitely by 2D experiments. Compounds 12 and 13 were new skeleton compounds that have never been reported. Meanwhile, the in vitro cytotoxic activities of the isolated compounds were assayed based on different human cell lines by MTT methods. In conclusion, this research has provided rational and experimential basis for the further development of this drug and quality control. |
