Objective:To observe the expression of IGFBP2 and ERK1/2 in serum samples and tumor tissues from patients with clear cell renal cell carcinoma(ccRCC)tissues and investigate the role and clinical significance of IGFBP2and ERK1/2 in the development and progression of ccRCC.Methods:Clinical data of 174 ccRCC patients who underwent surgical treatment in the Department of Urology,Affiliated Hospital of Jining Medical University from June 2017 to April 2019 were collected,and the blood samples of 30 healthy individuals were collected in the health examination center of our hospital during the same time period.The expression of IGFBP2 and ERK1/2 in ccRCC tissues and adjacent was assessed by immunohistochemistry and correlated with clinical stage,histological grade and tumour size;Serum concentrations of IGFBP2,ERK1 and ERK2 were also measured by ELISA in ccRCC patients and healthy controls.Results:The positive ratios of IGFBP2 and ERK1/2 expression in ccRCC tissues were higher(87.93%,6.67%;80.46%,10.00%)than those in adjacent noncancerous tissues,all of which were significantly different(χ~2=92.59,65.87;P<0.05).The positive ratios of IGFBP2 and ERK1/2 expression in tumors larger than 4 cm were higher than those in tumors smaller than 4 cm(93.94%,89.90%;80.00%,68.00%),all of which were statistically significant(χ~2=7.81,13.02;P<0.05).The expression of IGFBP2 and ERK1/2 did not differ statistically significantly by patients’age(χ~2=0.78,0.60),gender(χ~2=0.05,1.33),or tumor location(χ~2=0.96,0.57),(P>0.05).The positive expression rates of IGFBP2 and ERK1/2 increased with higher clinical stage and histological grade,and were statistically significant between groups in different grades(Z=-8.17,-3.80)and between groups in different stages(Z=-5.63,-9.94),(all P<0.05).ELISA showed that IGFBP2,ERK1,ERK2 were significantly increased in ccRCC patients(858.33±132.89μg/L;12.49±1.22μg/L;153.61±33.37ng/L)compared with healthy controls(267.65±86.56μg/L;8.69±1.80μg/L;105.63±12.03ng/L),all of which were significantly different(t=11.78,5.53,4.28;P<0.05).Conclusion:The expression of IGFBP2 and ERK1/2 in tumor tissue and adjacent tissue of ccRCC patients was analyzed by immunohistochemical staining,and it was found that the expression of IGFBP2 and ERK1/2 in tumor tissue was higher than that in adjacent tissue,and the difference was statistically significant(P<0.05).This indicates that both play a role in the occurrence and development of ccRCC.Secondly,the expressions of IGFBP2and ERK1/2 were correlated with tumor size,clinical stage,histological grade,etc.,and their expressions also showed an upward trend with the increase of stage and grade,and the difference was statistically significant(P<0.05).Correlation analysis showed that there was a positive correlation between the two in tissue expression(r=0.30,P<0.05),which indicated that the two may be related to the prognosis of ccRCC.Finally,it was found by ELISA that the serum levels of IGFBP2 and ERK1/2 in the ccRCC group were significantly higher than those in the healthy control group,which indicated that IGFBP2and ERK1/2 have a certain value in the diagnosis of ccRCC,and they play a role in the occurrence and development of ccRCC.It may play a synergistic role,which provides relevant ideas and methods for the diagnosis and treatment of ccRCC.
