| Objective:This study used liquid chromatography-mass spectrometry(LC-MS)and Western Blot to investigate the different expressions between renal carcinoma tissues and paracancerous tissues.Then we combined with Bioinformatics technology to analyze the relevant information,further explore the expression of col6a3 in renal cell carcinoma and its clinical significance.Method:1.Collecting the clinical samples which are confirmed renal carcinoma,then using LC-MS to construct differences in protein expression spectrum of renal carcinoma and find target protein COL6A3.2.Using Western Blot to verify the expression situation of COL6A3 in renal carcinoma and adjacent normal tissues.3.We explored four related database(STRING,Oncomine,COSMIC and c Bio Portal)to analyze information about the relationship between COL6A3 and renal carcinoma.Result:1.Using the liquid chromatography-mass spectrometry(LC-MS),21 differentially expressed proteins were obtained from renal cell carcinoma and its adjacent tissues,of which 14 were up-regulated and the remaining 7 were down regulated.The target protein COL6A3 was up-regulated.2.The results of Western Blot showed that the expression of COL6A3 in renal carcinoma tissues was significantly up-regulated compared with the adjacent tissues,which further verified the results of LC-MS.3.The analysis based on the STRING database found the interaction proteins of COL6A3.According to the analysis results,the main biological function of COL6A3 is platelet-derived growth factor binding,SMAD binding,protease binding,extracellular matrix structural constituent and integrin binding.It is mainly involved in negative regulation of endodermal cell differentiation,collagen biosynthetic and metabolic process,and platelet activation.The pathways are protein digestion and absorption,ECM-receptor interaction,and so on.The exploration on Oncomine databases showed that COL6A3 gene expression was up-regulated in renal carcinoma tissues.The analysis by searching COSMIC database indicated that COL6A3 gene can occur point mutation,copy number variation,gene expression and methylation in the kidney tissues,but the probability for variation was low.The proportion of missense substitution was the highest(62.03%),and the lowest proportion was inframe deletion(1.27%).The lowest proportion was in frame deletion(1.27%).By searching the cbioportal database,the coexpression genes of COL6A3 in renal cell carcinoma were found.According to the correlation,COL5A1 had the highest correlation.Conclusion:1.COL6A3 is an up-regulated expressed protein in renal cell carcinoma.2.Database analysis showed that COL6A3 can participate in the extracelluar matrix structural constituent,collagen biosynthetic and metabolic process.COL6A3 gene was up-regulated expression in renal carcinoma tissues,which was consistent with the protein expression in the experiment.Missense substitution is most likely to type for COL6A3 gene,which provides a new thinking for exploring the pathogenesis of the disease.In renal carcinoma,COL6A3 genes are the highest correlated with COL5A1 genes so that it can be used as the direction of further research.3.COL6A3 has close relationship with oncogenesis and tumor development,it may be a cancer-promoting gene for renal carcinoma.The clinical significance is that COL6A3 is expected to become an indicator of early diagnosis and screening of renal carcinoma,and can become a new marker to judge the prognosis of renal carcinoma. |
