Expression And Related Clinical Significance Of COL6A3 In Colorectal Cancer

Object:To screen the differential proteins of colorectal cancer and to investigate the expression of COL6A3 in colorectal cancer,and reveal its clinical significance at the same time.Methods:(1)The differential protein expression profile of colorectal cancer was constructed by using two-dimensional chromatography and mass spectrometry detecting the different proteins in the tissue samples of pathologically confirmed and resected colorectal cancer.(2)COL6A3 was selected as the target protein by consulting a series of literatures.Then the expression of COL6A3 were verified by Western blot,using four groups of colorectal cancer tissues and matched adjacent tissues samples.(3)The sera pecimens of definite diagnosis of colorectal cancer patients were collected as experimental group.Meanwhile,the sera of healthy physical examination patients collected as control group,which age was matched with colorectal cancer patients.Enzyme-linked immunosorbent assay(ELISA)was applied to detect COL6A3 expression in the two groups,and statistical approach was used to analyze the relationship between its expression with the patients clinical pathological parameters.Results:1.Through the analysis of liquid and mass spectrometry,44 different proteins were obtained in colorectal cancer tissue,of which 21 proteins were up-regulated in colorectal cancer tissue,and 23 proteins were down regulated in colorectal cancer tissue,and COL6A3 was one of the 21 up-regulated proteins.2.The results showed that the expression of COL6A3 in colorectal cancer tissues was significantly higher than that of the paracancerous tissues.3.(1)The mean value of COL6A3 concentration in serum of colorectal cancerpatients was significantly higher than that in the serum of healthy checkers,P<0.05.(2)In 42 patients with colorectal cancer,the serum COL6A3 concentration of colorectal cancer patients with middle differentiation was lower than that of low-differentiated colorectal cancer patients with the mean value of COL6A3 concentration,P<0.05.When the age,sex,lymph node metastasis and distant metastasis of colorectal cancer patients were separately analyzed,the mean difference of serum COL6A3 concentration in the two groups was not significant,P>0.05.Conclusions:1.The expression of COL6A3 in colorectal cancer was significantly higher than that in the paracancerous tissue.2.COL6A3 was significantly expressed higher in the serum of colorectal cancer patients,what's more,the expression level was connected with the degree of differentiation of colorectal cancer.Yet,it had no relationship with the gender,age,lymphatic metastasis,and distant metastasis of the patients.
