Robinia pseudoacacia L.is an important timber and protective tree species in my country.The genetic improvement research on the Robinia pseudoacacia L.families will help to promote the development of improved varieties of Robinia pseudoacacia L.and improve its economic benefits.In this study,46 half-sib families of Robinia pseudoacacia L.in Daqingshan Forest Farm,Feixian County,Shandong Province were used as the research objects,and the genetic variation of phenotypic traits was studied for their growth,shape,quality and seed setting,and the comprehensive evaluation of multiple traits was used to select elite families and elite singles.At the same time,the SSR molecular marker technology was used to analyze the genetic diversity of the excellent tree population of Robinia pseudoacacia L.,and the DNA fingerprint of the elite single plant of Robinia pseudoacacia L.was constructed,which laid a theoretical foundation for the future research on genetic improvement and molecular breeding of Robinia pseudoacacia L..The main findings are as follows:(1)From the phenotypic level,this study conducted genetic variation research on growth traits,morphological traits and fruit-setting traits of 46 half-sib families.The coefficient of variation was between14.61% and 48.28%,and the heritability of most of the traits was greater than 0.5.The rich variation and high heritability were conducive to the selection of elite families.(2)In this study,46 Robinia pseudoacacia L.half-sib families were comprehensively evaluated for multi-characters in terms of growth traits and morphological traits,and 5 elite families were selected: F41,F23,F44,F12,and F2.96 elite trees were selected based on indicators such as morphological and morphological indicators;the selected elite families and individual trees had obvious genetic gain.Through selection in terms of fruit-bearing characteristics,7 families with elite results were selected: F22,F17,F19,F25,F27,F37,F44,which can be used as an alternative material for improving seed gardens.(3)At the molecular level,10 pairs of SSR primers were used to analyze the genetic diversity of Robinia pseudoacacia L.at the molecular level in this study,and a total of 93 allelic loci were detected,with an average locus of 9.3;each pair of SSR polymorphisms The sexual information content was all greater than 0.5,and the average expected heterozygosity(0.700)was higher than the observed heterozygosity(0.672).The selected Robinia pseudoacacia L.tree has high genetic diversity and can be used as a candidate material for Robinia pseudoacacia L.seed garden.Population genetic structure analysis divided 92 trees into 5 subgroups;the results of AMOVA showed that the genetic variation of Robinia pseudoacacia L.population was mainly derived from inter-individual variation.(4)In this study,the combination of the 10 selected primer pairs can completely distinguish92 trees,and the DNA fingerprints of the 92 trees were constructed at the molecular level.Highefficiency molecular markers of trees;RP07 and RP150 are high-efficiency molecular markers that can distinguish polyploidy of Robinia pseudoacacia L.,and play an important role in the study of polyploidy of it.This primer combination and fingerprint map can be used for the molecular identification of Robinia pseudoacacia L.families and the selection and breeding of elite hybrid combinations in the future. |
