In this study,the F1population of male sterile germplasm‘JMS2’×wild sour jujube superior line‘Xing16’was used as test materials to determine the sugar and acid components and quality traits of fruit of hybrid offspring,and to explore the genetic law of fruit sugar and acid traits of hybrid offspring.Combined with the constructed high-density genetic map of jujube,QTL mapping for related quality traits was carried out.The results were as follows:(1)The contents of fructose,glucose,sucrose,oxalic acid,citric acid,malic acid and succinic acid in fruit of hybrid progenies of jujube conformed to the genetic characteristics of quantitative traits.In the two years,the variation range of fructose,glucose and sucrose was 31.80%~38.65%,32.05%~38.06%and 25.64%~29.60%,and the average fructose content was 7.54%and 6.46%,while the average glucose content was 8.19%and 7.65%,which were lower than the median value,showing a declining trend.The average sucrose content was 18.90%and 19.75%higher than the median value.The variation range of oxalic acid,malic acid,succinic acid and citric acid content in the two years was 30.37%~50.39%、44.80%~48.26%、52.08%~69.19%and 67.58%~76.77%.The average content of oxalic acid was 0.09%and 0.07%,the average content of succinic acid was 0.17%and 0.25%,the average content of citric acid was 0.04%,and the average content of malic acid was 0.33%,which was lower than the median value,showing a declining trend.Only in 2021,the average content of malic acid was 0.31%higher than the median value.(2)QTL localization of fruit sugar content traits was carried out to obtain 29 sites,including 6fructose QTL loci,9 glucose QTL loci,14 sucrose QTL loci,and 9 principal QTL loci;fructose and glucose have a QTL loci coincidence.(3)Fruit acid content traits were localized to a total of 66 QTL sites,15 citric acid QTL sites,17malic acid related QTL sites,24 oxalic acid related QTL sites,and 6 succinic acid related QTL sites.A total of 29 main effect QTL sites were detected for acid content traits.The largest number of QTL sites are located on the chain group LG1,reaching 19.Citric acid was detected at 1 site of coincidence with oxalic acid,and citric acid was detected at a site of coincidence with malic acid.(4)Location of nutrients in fruit of hybrid offspring of jujube.A total of 109 QTLs were identified for fruit nutrients in jujube hybrids,including 8 QTLs related to total sugar,29 QTLs related to total acid,34 QTLs related to total flavonoids,25 QTLs related to total phenols,9 QTLs related to protein,and 4 QTLs related to VC.A total of 37 major QTL loci were detected in fruit internal nutrients,and no major QTL loci were detected in protein and VC content traits.The most QTLs were located on linkage group LG6.Total flavonoids and total phenols,total acids and VC were all located at the same site,and total acidic traits were repeatedly detected at the same site in two years,which could be stably inherited.
