Muscovy duck is an excellent special meat poultry,which has the advantages of fast growth,tolerance to coarse feeding and less feed consumption.As the reproductive organ of male animals,testis development is closely related to reproductive performance.Studies have shown that arginine,astragalus membranaceus,epimedium and fructus psoraleae can promote testis development,but few studies on these drugs in Muscovy duck testis.A total of 150 49-week-old male Muscovy ducks were randomly divided into five groups with three replicates per group and ten ducks per replicate.Group A was the control group and fed a basal diet.Group B,C,D and E were fed the basal diet supplemented with 0.4% arginine,1% astragalus membranaceus,1% epimedium and 1% fructus psoraleae,respectively.After75 days of feeding,histomorphological observation,blood biochemical indexes detection,and transcriptome sequencing technology were used to sequence the testis,and the main results were as follows:(1)The testicular histomorphology results showed that compared with group A,the number of spermatogenic epithelium and spermatogenic cells and mature sperm were increased in group B,C,D and E,which improved the morphology of testis tissue.(2)The results of serum reproductive hormone showed that compared with group A,the contents of T,E2,FSH and LH were increased in group B,C,D and E,and those in group D and E were significantly higher than those in group A(P<0.05).(3)The results of serum antioxidant indexes showed that compared with group A,the contents of SOD,GSH-Px and T-AOC were significantly increased in group B,C,D and E(P<0.05),while the content of MDA was significantly decreased(P<0.05).(4)Lnc RNA library was constructed and sequenced,in four combinations of A vs B,A vs C,A vs D,A vs E were identified in 1268(740 up-regulated,528 down-regulated),909(447up-regulated,462 down-regulated),2414(1538 up-regulated,876 down-regulated),1523(849 up-regulated,674 down-regulated)differentially expressed m RNAs;756(305 upregulated,451 down-regulated),536(247 up-regulated,316 down-regulated),1406(453 upregulated,953 down-regulated),and 776(319 up-regulated,457 down-regulated)differentially expressed lnc RNAs.Combined with GO and KEGG enrichment analysis,MAPK signaling pathway,PI3K-Akt signaling pathway,c AMP signaling pathway and other signaling pathways related to testis development were significantly enriched.Among them,genes related to the signaling pathways include PPP3 CA,CDC25B,FGF10,LNC_026244,LNC_002321,LNC_002322,etc.(5)Small RNA library was constructed and sequenced,in four combinations of A vs B,A vs C,A vs D,A vs E were identified in 50(34 up-regulated,16 down-regulated),34(16 upregulated,18 down-regulated),225(120 up-regulated,105 down-regulated),169(64 upregulated,105 down-regulated)differentially expressed mi RNAs.Combined with GO and KEGG enrichment analysis,Notch signaling pathway,TGF-β signaling pathway,Wnt signaling pathway and other signaling pathways related to testis development were significantly enriched.Among them,the genes related to the signaling pathways include mmu-mi R-205-5p,has-mi R-139-5p,mmu-mi R-139-5p,etc.In conclusion,0.4% arginine,1% astragalus membranaceus,1% epimedium and 1%fructus psoraleae can promote testis development of Muscovy duck,and participate in the signaling pathways related to the testis development and the differential expression of pathway genes.
