| Salmonella abortus equi(S.abortus equi)is the pathogen of horse of infectious disease dominated that cause horse abortion with the rapid development of horse and donkey industry and intensive breeding,the disease is become serious problem.Vaccine is an important measure to prevent the disease,but effectiveness of different vaccines of different vaccines are quite different.In this studying mouse and horses as research models,were compare the immune effects of three vaccines and analyze.The result will provide basis of horse Salmonella vaccine selection.The mice or horse were immunized with inactivated vaccine,attenuated vaccine and Flj B and Fli C proteins.The antibody level,efficacy and antibody subtype of mice and horses were detected by indirect ELISA method.The results showed that the antibody level of each immunized group were significantly higher than those of control immune groups(P < 0.05),and the antibody effect was up to(1:218,700).The antibody subtypes produced by attenuated vaccine and inactivated vaccine are Ig G1,Ig G2 a and Ig G2 b,antibody subtype immunized with Flj B and Fli C are mainly Ig G1 and Ig G2 b.The profection test results of immunized mice showed that the protection rate was 87.5% in Attenuated H,Attenuated M and Inactivated H groups,75% inactivated group,inactivated M group,Flj B group and Fli C and Flj B co-immunization group was and 62.5% in Fli C group.The body weight of group began to regain in the 3d to 4d after the attack,and the bacteria vaccine in the liver,spleen and kidneys of each immune group was significantly lower than that of PBS control group(P< 0.05),the Attenuated H immunization group was the lowest.then followed by the Inactivated H group,The pathological results of liver,spleen and kidney of mice in each experimental group showed that the lesions in each dose group of attenuated vaccine and inactivated vaccine were improved compared with the PBS group.The results of horse antibody assay showed significantly higher antibody level were indaced,in the attenuated H group and Inactivated H group than other groups(P < 0.05),and the levels of Fli C and Flj B combined immune antibodies higher than those of protein alone immunity;were the main subtypes of by each immune group,antibody effect induced by inactivated vaccine and attenuated vaccine was up to(1:72,900),which was higher than that of subunit vaccine and combined vaccine group(1:14,300).Antibody analysis results showed that the Ig G subtypes produced by each immune group mainly were Ig G1 and Ig G2 b.To sum up,mice and horses are immunized with 3 different vaccines,both attenuated and inactivated vaccines can produce higher antibody levels than immunized Fli C and Flj B proteins,reduced the amount of they organs bacteria load and tissue damage in mice,and the immune effect of inactivated and attenuated vaccine was affected by the dose. |
