Cancer is one of the most deadly diseases in the world. The rapid and accurate detection of cancer cells is of great significance to basic research, clinical diagnosis, and early treatment.Metal-organic frameworks(MOFs)because of their large specific surface area, pore, and other excellent features have been successfully applied to various sensors. The detection of cancer cells is one of its important application directions. The test uses the biosensor to realize the rapid detection of U937.1)Firstly,a glassy carbon electrode(GCE)was selected and pretreated, and multi-walled carbon nanotubes(MWCNTs)and copper-based metal-organic framework(Cu-MOF)were sequentially modified on the surface of GCE to form Cu-MOF/MWCNTs/GCE.Then the surface of the sedimentary layer is modified. An Ab/SA/Cu-MOF/MWCNTs/GCE sensor was constructed by using the affinity between streptavidin(SA)and biotin to orientate biotinylated Ig G3 antibody to the surface of the sensor.2)The sensors modified by MWCNTs and Cu-MOF in the sequence were characterized by cyclic voltammetry(CV),electrochemical impedance spectroscopy(EIS),scanning electron microscopy(SEM),infrared spectroscopy Raman and X ray diffraction. And their electrochemical properties, morphological structure, and components were analyzed. The results of characterization analysis showed that the Cu-MOF electrochemical cell sensor was successfully constructed.3)Sensors with different encapsulation levels were used to detect 5 gradient concentrations of cells. The equivalent circuit was used to fit the tested Nyquist plots, and the fitted data were used to further analyze the relationship between each equivalent element and the detection sensitivity. The research results showed that the electrochemical cell sensor based on MOFs could easily detect cancer cells within the range of 3×10~1~3×10~4cells/m L,and the detection limit of the sensor was 30cells/m L.At the same time, the sensor has good specificity, stability and repeatability, which provides a new method for clinical detection of cancer cells.Figure 36;Table 12;Reference 69...
