In this paper,we analyzed 99 kinds of ornamental pepper.These ornamental pepper provided by the Solanaceous fruit laboratory of Vegetable research institute of Guangdong academy of agricultural sciences.We research about 37 agronomic traits of plants and using diversity analysis、cluster analysis and correlation analysis these way to processing the data.To understand the classification and the relationship between pepper germplasm resources.We constructing the 6-generation population(P1、P2、F1、BC1、BC2、F2)of the hybridized combination of purple and white pepper.Use visual observation research genetic law by visual observation of pepper about color of petals and the unripe fruit.And understand the genetic pattern of the petal color and fruit color(purple/white)of the experimental pepper.We used BSA to analyze F2.Sequencing DNA from two extreme pools and two high-quality genomes of parents and use QTL-seqr to calculate the Gprime and △ SNP index of two pools of F2 population.According to the result of calculation,the purple gene of pepper was located at the position of related chromosome.Next,develop molecular markers within the interval and detect in recessive individual strains to analyze whether the marker is linked to the target gene.That we can verify the results of the location.This study mainly obtained the following results:1.The average variation coefficient of 37 agronomic traits of 99 ornamental pepper germplasm resources was 43.07%.The highest agronomic traits is 128.50% of fruit umbilical appendage and the lowest was 12.77% of style length.The average shannon-weaver diversity index is 1.095,which the highest diversity index is 2.388 and the lowest is 0.349.When the Euclidean distance is 10,We can divide 99 ornamental pepper ingredients into 2 categories,and when is 5,It can be into 4 categories and It is based on cluster analysis of 37 agronomic traits.In the correlation analysis,the agronomic traits such as plant height,plant width and branching were significantly correlated with several agronomic traits.In this experiment,99 ornamental pepper germplasm resources had rich variation degree and diversity.According to the analysis results,the target traits can be improved.We can accelerate the innovation and breeding of ornamental pepper.2.When the white pepper and purple pepper are crossbreeding,The results of genetic law analysis showed that the color of petals and fruits of pepper were controlled by a pair of nuclear genes.The gene that controls purple has an superimposed effect,Purple is dominant to white.3.We used BSA-seq technology to localize purple gene of ornamental pepper.The gene range of the ornamental pepper purple trait was rapidly and accurately reduced to a4.143 mb section of chromosome 10(chr10:159881514-164044093).Then In Del primers were designed for the initial localization region of the gene.The exchange rate of the plants was calculated based on the amplification results of F2 and BC1 populations,and then the linkage between the three markers(15996-1、16101、16367)and the target gene could be inferred.In this way,the accuracy of the location area can be further verified.In this study,we lay a foundation for continue to do fine gene localization and molecular marker assisted breeding of purple pepper. |
