| Inner Mongolia cashmere goat is an important cashmere goat breed in China,and its cashmere is an important wool textile raw material,which has the advantages of fineness,softness,lightness and so on.Cashmere is a kind of keratinized product derived from the secondary hair follicles of cashmere goats.the growth cycle of secondary hair follicles of Inner Mongolia cashmere goats can be divided into th ree stages: growth period,retrogression period and resting period.the process of growth and shedding of cashmere is directl y related to a series of cyclical biological changes from active proliferation to apoptosis in the secondary hair follicle and then to the cessation of growth in the resting period.It is found that many 1nc RNA grow periodically in skin hair follicles,so in this study,transcriptome sequencing was carried out on the skin tissues of cashmere in different months by RNA-seq technique,and the data of three periods of skin hair follicle growth,retrogression and resting were compared,and significant differentiall y expressed 1nc RNA and genes were obtained and anal yzed by bioinformatics,and finall y verified by q RT-PCR.The potential 1n RNA that may regulate the cycle transition of skin hair follicles was screened to provide a theoretical basis for further study of the function of1 nc RNA and the role of specific 1nc RNA in the cycle transition of skin hair follicles.The specific results are as follows:1.A total of 5138 reliable 1nc RNA transcripts were identified in the goat skin sample library in ten months,including new lnc RNA1299 and known lnc RNA3839,including 4022 intergenic 1n RNApredictor 543 antisense 1n RNA1 nc RNA exons 461.The ide ntified 1nc RNA had fewer exons,shorter transcript length,lower expression and shorter open reading frame.2.5157 transcripts and 275 lnc RNA were differentiall y expressed in hair follicles between resting and growing skin.2265 genes were up-regulated and2892 down-regulated.Lnc RNA192 was up-regulated and 83 down-regulated.There were 5157 genes and 378 lnc RNA differentiall y expressed in the growth phase and the retrogression phase,2846 genes were up-regulated and 2311 genes were down-regulated.Lnc RNA316 was up-regulated and 62down-regulated.There were 5042 genes and 478 lnc RNA differentiall y expressed in retrogression and resting phase,and 2598 m RNA transcripts were up-regulated and 2444 down-regulated.Lnc RNA420 was up-regulated and 58down-regulated.3.Partial differential transcripts and lnc RNA target genes were enriched in biological processes such as hair follicle cell migration,capillary regeneration,cell signal transduction and regulation,as well as VEGF and MAPK signal pathways related t o cell proliferation.Some differential transcripts and lnc RNA target genes are enriched in biological processes such as DNA double-strand unwinding,negative regulation of translation,keratin filaments and apoptosis-related PI3K-Akt signal pathways.Part ial differential transcripts and lnc RNA target genes were enriched into biological processes such as cell component decomposition,organogenesis,cell cycle regulation and Rap1 signal pathways related to Cellcycle and apoptosis.4.The differentiall y expr essed 1nc RNA and genes in 12 different periods were verified by q RT-PCR,and the results showed that the trend of q RT-PCR results was consistent with that of RNA-seq sequencing.Combined with literature anal ysis,some key lnc RNA and its target genes pairs such as MSTRG.528.1(FGF21),MSTRG.25611.2(VEGFA)which may be related to skin hair follicle cycle transition were screened. |
