| MAPK signaling pathway can induce the proliferation and differentiation of hair follicle cells and promote the periodic development of hair follicles,and then affect the growth of hair,the distribution of hair follicles and the number of hair trunks.TNFRSF1A,one of the important members of the MAPK signaling pathway,is of great significance for the periodic growth of cashmere.In this study,qPCR and Western blot were used to detect the changes of TNFRSF1A in the hair follicle cycle of skin.RNA interference technology was used to explore the effects of TNFRSF1A expression on the upstream and downstream gene expression of TNFRSF1A in the MAPK signaling pathway.The results are as follows:1、qPCR results showed that the expression of TNFRSFIA in Inner Mongolia cashmere goats was the highest in May and the lowest in October,and the expression level in Anagen was significantly lower than that in Catagen and Telogen.We speculate that the high expression of TNFRSFIA in May is an important factor leading to cashmere shedding in Inner Mongolia cashmere,while in October,the expression of TNFRSFIA reached the lowest level in the flourishing period of cashmere growth.2、Western blot results showed that the expression of TNFRSF1A was the lowest in hair follicle Anagen(October),followed by hair follicle Catagen(December),and the highest in hair follicle Telogen(March)and hair follicle Early Anagen(May).The key period of reconstruction of skin follicles in cashmere goats is around May,and the flourishing period of villus growth in October.This result further speculate that TNFRSF1A is related to cashmere shedding.3、By transfecting shRNA into fetal fibroblasts of Inner Mongolia cashmere goats,the expression of TNFRSF1A was detected by qPCR and Western blot.The results showed that the expression of TNFRSF1A in the transfected group was significantly lower than that in the blank control group,and the expression of TNFRSF1A in shRNA-2 was lower than that in shRNA-1 and shRNA-3.Based on the successful interference of TNFRSFIA,the expression of upstream and downstream genes in MAPK signaling pathway was analyzed.qPCR results showed that the expression of TNF and TRAF2 was significantly lower than that of negative control group,while the expression of TRADD,TAB1 and TAK1 was significantly higher than that of negative control group.4、Prediction maps of TNFRSF1A and PPI were constructed by STRING website,and the transduction process of TNFRSF1A in cashmere goat MAPK signaling pathway was successfully expounded. |
