In this test,three batches of Swine Foot-and-Mouth Disease Type O and A Bivalent Inactivated Vaccine were prepared and tested,according to the Manufacturing and Inspection Procedures of Swine Foot-and-Mouth Disease Type O and A Bivalent Inactivated Vaccine.Seed virus prepared by swine vesicles,neonatal rat,basic cell and virus solution for vaccine of preparation was qualified.After antigen solution was inactivated,purified and concentrated,it was mixed with adjuvants to prepare three batches of vaccine.The endotoxin content of the three batches of vaccine was less than0.78 EU per dose,and the total protein content was 48-56μg per dose,and total 146 s virus content was 14-18μg per dose,and all the quality indicators of the product were higher than the quality standards stipulated in the regulations.The vaccine was also safe for guinea pigs,mice,and pigs.According to the Manufacturing and Inspection Procedures of Swine Foot-and-Mouth Disease Type O and A Bivalent Inactivated Vaccine,the efficacy test of three batches of vaccine was divided into 3 dose groups of 1 dose(2m L),1/3 dose(0.67 m L),1/9 dose(0.22 m L),and the PD50 values were all 15.59,which were calculated using the Reed-Muench method on the basis of the number of protection of the immunized pigs.In addition,the other efficacy test of high-efficiency vaccine was divided into 3dose groups of 1/2 dose(1m L),1/6 dose(0.33 m L),1/12 dose(0.17 m L)and the othe r 3dose groups of 1/4 dose(0.25 m L),1/12 dose(0.17 m L),1/36 doses(0.06 m L),and the PD50 values were 31.18~50.05,which were calculated using the Reed-Muench method on the basis of the number of protection of the immunized pigs.By selecting the new dose group to determine the final effectiveness of the prepared vaccine,it can guide the production enterprises to add and control the antigen amount,or effectively guide the immunization of foot-and-mouth disease vaccine.
