| The complete elimination of dormancy of flower bud is one of the important conditions to ensure the flowering quality of peony.This article material,peony flower bud of peony flower bud dormancy breaking three periods in the process of the transcriptome sequencing,then pick a plays an important role in the peony flower bud dormancy breaking of ERF transcription factor genes,by virus induced gene silencing technology preliminary validation of the gene function,gene cloning and bioinformatics analysis,including expression pattern analysis and function research.Theresults are as follow:1.After silencing PsERF11 gene by VIGS technology,the growth rate of peony was significantly slowed down.After 13 days of infection,PsERF11 silenced plants were in the state of just germination,while TRV no-load control plants were in the state of beginning elongation.At 24 days after bacterial fluid infection,the stem of PsERF11 silenced plants began to elongate and the leaves were slightly expanded,while the stem of TRV no-load control plants was longer and the leaves were expanded.The results showed that PsERF11 silenced plants grew significantly slower than TRV no-load control plants.2.The expression levels of PsERF11 gene in the VIGS silenced peony plants were lower than TRV no-load control,the expression levels of PsGA3ox1 a in the PsERF11 silenced peony plants were significantly lower than TRV no-load control,and the expression levels of PsNCED2 and PsAAO in the PSERF11 silenced peony plants were significantly higher than TRV no-load control.3.Dual luciferase assay results showed that PsERF11 could promote the activity of promoter of PsNCED2,a structural gene of ABA biosynthesis pathway,but had no significantly effect on the activities of PsAAO and PsGA3ox1 a.4.An ERF transcription factor gene named PsERF11,was cloned from leaves of tree peony and related to dormant release of flower buds.The complete CDS sequences was 369 bp,encoding 122 amino acids,belonging to ERF transcription factor,with typical conserved domain and functional domain of ERF family. |
